specific Embodiment approach 1

[0029] A method for the stable germination of conidia of tomato gray leaf spot and for the identification of artificial inoculation at the seedling stage, comprising the following steps:

[0030] Step a, prepare gray leaf spot bacteria spore suspension;

[0031] Step b, artificial inoculation: Spray the spore suspension prepared in step 1 directly on the front and back of the leaves at the tomato seedling stage, grow in the dark, and after 3 to 5 days, screen tomato materials resistant to gray leaf spot.

[0032] A kind of screening method of gray leaf spot of tomato described in the present embodiment, step a prepares the method for the spore suspension of gray leaf spot bacteria comprising the following steps:

[0033] Step a1. Treatment of tomato leaves: select fresh tomato leaves without disease spots, rinse them with tap water first, then soak them in alcohol for 5-10 seconds, put them in mercuric chloride solution for 1 minute, and then rinse them with sterile water for ...

Abstract

A method for stable germination of conidia of tomato gray leaf spot pathogens and for artificial inoculation identification in a seedling stage belongs to the field of prevention and control of tomatogray leaf spot pathogens. The invention aims to solve the problem that the spore germination efficiency of the tomato gray leaf spot pathogens is low and not suitable for spore production. The methodprepares a spore suspension of the gray leaf spot pathogens and then conducts artificial inoculation by directly spraying the prepared spore suspension on front and back sides of the leaves in the tomato seedling stage, growing the tomatoes away from light for 3 to 5 days, and screening tomato materials resistant to the gray leaf spot disease. According to the method, a PDA medium is inoculated with purified tomato gray leaf spot pathogen S. lycopersici strains, and the strains are cultured for 8-12 days; and then the pathogens on the medium are picked and placed on sterile tomato leaves, andcultured for 1 to 2 weeks. The method can significantly improve the germination rate of the conidia of the tomato leaf spot pathogens, and establish a simple and stable technical system to improve the germination rate of the conidia of the tomato pathogens.

Description

technical field [0001] The invention belongs to the field of tomato gray leaf spot prevention and control, and in particular relates to a method for the stable germination of tomato gray leaf spot conidia and its use for artificial inoculation identification at the seedling stage. Background technique [0002] Tomato Gray Leaf Spot (Tomato Gray Leaf Spot) is mainly caused by the infection of S. lycopersici and S. solani S. solani. The leaves with gray leaf spot disease were collected from Chifeng in Inner Mongolia, Shouguang in Shandong, and Harbin in Heilongjiang for separation and purification. It indicated that the main species of local pathogen in Harbin was Stemphylium lycopersici, not Stemphylium solani. The disease occurs worldwide, but is severe in warm and humid regions. In recent years, tomato gray leaf spot has become a new disease that endangers tomato production in many provinces of my country, and has caused serious harm to tomato production. At present, the...
