Abstract

The invention discloses an RNA interference method based on a bemisia tabaci nymph body wall permeation approach. The method comprises the following steps of (1) cultivation of cotton seedlings; (2) insect inoculating and spawning; (3) RNA interference based on the body wall permeation approach, wherein dsRNA and an exogenous control gene dsEGFP are prepared, the dsRNA is uniformly mixed with a nano material of a star-shaped polymer according to the volume ratio of 1:1, the mixture is carefully dropped on nymphs, cotton leaves are inserted back into a bottle filled with clean water after the liquid drops are absorbed by the insects, the insects are placed into a culture box for continuing feeding, the death number is recorded 2 days later, and nymphs samples are collected. According to themethod, by utilizing the strong permeability of the novel nano material, the dsRNA quickly permeates into bemisia tabaci bodies through the body walls, so that the death rate is obviously reduced, the interference efficiency is improved, and the timeliness and the repeatability are high. In addition, the use amount of the dsRNA in the method is very small and reaches about 0.3 microliter per individual, the interference efficiency is ensured, and meanwhile the experimental cost is greatly reduced; the method has a good application prospect.
