[0046] Example 1 Prokaryotic expression and purification of the mature peptide of the anti-inflammatory and anti-thrombotic polypeptide Motistin of Myotis brucei

[0047] 1. Construction of prokaryotic expression system

[0048] The prokaryotic expression system was constructed by Beijing Qingke Xinye Biotechnology Co., Ltd. The nucleotide sequence of the mature peptide of Motistin (as shown in SEQ ID NO: 3) was synthesized and connected to the Pet-32a recombinant plasmid, and finally transformed into into BL21 strain.

[0050] 1) Cell collection

[0051] After the obtained carrier bacteria liquid was plated to obtain a single clone, the single clone was picked and sequenced to verify the correctness of the carrier sequence, and the monoclonal bacteria were frozen at a certain concentration of glycerol.

Abstract

The present invention provides a mature peptide of Motistin, an inhibitor of leukotriene A4 hydrolase in Myotis brucei, and its application, belonging to the technical field of biomedicine; the amino acid sequence of the mature peptide is shown in SEQ ID NO:2. The present invention finds for the first time that the mature peptide of Motistin can inhibit the hydrolase activity of LTA4H without affecting the aminopeptidase activity thereof. The mature peptide of Motistin can target LT44H, inhibit the recruitment of immune cells and the production of inflammatory mediators by inhibiting the hydrolase activity of LTA4H, and also inhibit the production of inflammatory cytokines IL6, TNFα, IL1β caused by viruses and bacterial lipopolysaccharides , has the potential to be an effective drug for inflammation suppression.

Description

technical field [0001] The invention relates to the technical field of biomedicine, in particular to a mature peptide of Motistin, an inhibitor of leukotriene A4 hydrolase of Myotis brucei Brycei, and an application thereof. Background technique [0002] LTA4H is the final and rate-limiting enzyme of the arachidonic acid pathway. It is known for its epoxyhydrolase function, converting LTA4 to LTB4 (Dennis, E.A. & Norris, P.C. Eicosanoid storm in infection and inflammation. Nat Rev Immunol 15, 511–523 (2015)). [0003] LTB4 is an important pro-inflammatory lipid mediator that initiates and amplifies innate and adaptive immune responses and is chemoattractive to immune cells and releases inflammatory mediators (Peters-Golden, M. & Henderson, W.R. Leukotrienes. NEngl J Med 357, 1841–1854 (2007); DiGennaro, A. & Haeggstrom, J.Z. Theleukotrienes: immune-modulating lipid mediators of disease. Adv Immunol 116, 51-92 (2012)). Engagement of its receptors BLT1 and BLT2 has particula...
