Abstract
The invention discloses a mutagenesis method of a seed lotus phenotypic variation mutant plant. The mutagenesis method comprises the following steps of (1) shelling seed lotus seeds; (2) soaking the shelled lotus seeds in clear water to accelerate germination, and then cutting off part of cotyledons to expose germ growing points; (3) carrying out mutagenesis treatment by using an ethyl methanesulfonate solution as a mutagenic agent; and (4) performing rooting and germination in water, then performing field planting, and obtaining the mutant plant with phenotypic variation through phenotypic observation. A lotus EMS mutation system is explored by taking the seed lotus seeds as a material, and a result shows that shells of the lotus seeds are completely removed, so that the lotus seeds swelland germinate quickly and germinate orderly and consistently; and the shelled lotus seeds are soaked in clear water for 24 hours, 1/2-1/3 of the cotyledons are cut off, and the cotyledons are treatedin an EMS solution with the concentration of 0.5% for 16 hours to obtain mutant materials with various phenotypes, thereby providing a new germplasm material for seed-provided lotus breeding and effectively solving the problem of genetic stenosis of seed-provided lotus.
