Problems solved by technology

However, the above-mentioned markers need to sequence the PCR products, which is cumbersome and low-throughput, and there is still a certain distance between the two markers and the target gene.
The application of these markers in watermelon fruit firmness molecular marker-assisted breeding has certain limitations

Embodiment 1

[0065] Example 1, the development of SNPs related to the firmness of watermelon pulp, the development of Hf-KASP1 markers and the establishment of genotype detection methods

[0066] 1. Test materials

[0067] The test materials include paternal parent, female parent, BC 1 f 9 group;

[0068] The male parent is: GS43 (PI482307, https: / / npgsweb.ars-grin.gov / gringlobal / accessiondetail?id=1377243), which is a typical wild type watermelon material with high fruit hardness (the flesh hardness value is 15Kg / cm 2 ), low sugar content;

[0069] The female parent is: TWF, which is a typical East Asian type of cultivated watermelon variety, and is an excellent parent material for commercial hybrid melons. It has high sugar content, but the flesh is brittle and easy to crack (the hardness value of the flesh is 5Kg / cm 2 ).

[0070] BC 1 f 9 Groups are: BC above 1 f 8 BC 1 f 9 Group, a total of 144 strains

[0071] All the above-mentioned test materials are germplasm resource m...

Embodiment 2

[0124] Embodiment 2, verification of KASP1 marker

[0125] In order to further verify the linkage relationship between the Hf-KASP1 marker in Example 1 and the watermelon pulp firmness gene Hf (corresponding trait: pulp firmness), natural populations were used for verification. Specific steps are as follows:

[0126] The test material is a natural resource group; the natural group is: a natural group composed of 96 representative watermelon germplasm resources in 1484 watermelon resource banks (see Table 2 for details); each test material in Table 2 is a Beijing Germplasm resource materials preserved in the Germplasm Resource Bank of the Vegetable Research Center of the Municipal Academy of Agriculture and Forestry Sciences.

[0127] With reference to Example 1, on the one hand, each watermelon germplasm resource for testing was tested for pulp hardness (obtaining the value in the "hardness" column in Table 2), and on the other hand, the Hf-KASP1 marker was used to identify t...

Abstract

The invention discloses an SNP (Single Nucleotide Polymorphism) site for judging the hardness of watermelon pulp, an Hf-KASP1 marker and application of the SNP site and the Hf-KASP1 marker. The invention provides an application of a substance for detecting whether 13317084th deoxyribonucleotide on a chromosome 6 in a watermelon genome is T or A or T and A in any one of the following aspects: identifying the hardness of watermelon pulp, and breeding a hard-pulp / non-hard-pulp watermelon variety. On the basis of a watermelon 97103 whole genome sequence and re-sequencing information of a hard-flesh material GS43 (PI482307) and a cultivated soft-flesh material TWF, according to a determined SNP design marker, a BC1F9 generation positioning group obtained after hybridization of a wild hard-flesh watermelon GS43 and the cultivated watermelon TWF is utilized to carry out fine positioning on a watermelon hard-flesh gene, and a closely-linked KASP marker is developed. The molecular marker KASP can be used for initial screening of watermelon pulp hardness varieties, and the purpose of molecular assisted breeding is achieved.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to a SNP site for judging the firmness of watermelon pulp, a Hf-KASP1 marker and an application thereof. Background technique [0002] Watermelon (Citrullus lanatus) is an important vegetable crop in my country, and its production and sales rank first in the world. With the development of fruit production, sales and transportation channels, watermelon fruit firmness, as an important watermelon quality trait, has been paid more and more attention by people. [0003] At present, the market favors watermelon varieties with hard flesh, which are resistant to storage and transportation and are convenient for fruit slicing and plattering. However, most traditional cultivars have crispy flesh. How to effectively cultivate hard flesh varieties is currently a hot spot in watermelon breeding. Breeding practice has proved that the use of molecular marker-assisted breeding can greatly shorten the ...
