[0119] Example build beta 2 m knockout chimeric mice

[0120] 4-week-old female C57B1 / 6J (β 2 m + / + ) and C57B1 / 6J-β 2 m tm1 / Unc (β 2 m - / - ) mice were purchased from Jackson Laboratory (Bar Harbor, ME). beta 2 m + / + and beta 2 m - / - Both are exchanged for bone marrow transplantation. The preparation of bone marrow chimeras is described in detail in Sobel et al., J. Exp. Med., 1991, 173:1441, which is incorporated herein by reference in its entirety. Briefly, recipient mice (β 2 m + / + and beta 2 m - / - ) lack of NK cells. This pretreatment prevents radioresistant NK cells in C57B1 / 6J mice from responding to C57B1 / 6J-β-derived 2 m tml / Unc The bone marrow cells of the mice were rejected. On the day of interchange bone marrow transplantation, recipient mice were lethally irradiated in two equal doses, 3 hr apart, for a total of 1050 rads. Donor mice were sacrificed and bone marrow was collected by flushing the tibia and femur. Bone marrow cells were incubated ...

Abstract

Methods of inducing an immune response against an immunogen in an individual are disclosed. The methods comprise administering to the individual, one or more nucleic acid molecules that comprise a nucleotide sequence that encodes an immunogen and a nucleotide sequence that encodes a Major Histocompatibility Complex antigen. The nucleotide sequences that encode the immunogen and Major Histocompatibility Complex antigen are expressed when taken up by cells of the individual and an immune response against the immunogen is induced in the individual. Methods of reducing rejection of unmatched donor cells, tissue or organ in an individual undergoing cell, tissue or organ transplantation are disclosed. The methods comprise administering to the individual, one or more nucleic acid molecules that comprise a nucleotide sequence that encodes a death signal or toxin and a nucleotide sequence that encodes a major Histocompatibility Complex antigen that is matched to the donor cells, tissue or organ. The nucleotide sequences that encode the Major Histocompatibility Complex antigen and death signal or toxin are expressed when taken up by cells of the individual. T cell death through interaction with the death signal or toxin results in a reduction of rejection of unmatched donor cells, tissue or organ. Methods of reducing a dominant immune response in an individual and methods of expanding a subpopulation of T cells associated with a specific immune response are also described. Plasmids and compositions comprising plasmids useful for practising the method are described.

Description

field of invention [0001] The present invention relates to vaccines and methods for inducing a prophylactic and / or therapeutic immune response in an individual. The present invention relates to compositions and methods for treating patients undergoing cell, tissue and / or organ transplantation. The present invention relates to compositions and methods for modulating an immune response. Background of the invention [0002] Vaccines can be used to immunize an individual against a target antigen, such as a pathogen antigen or an antigen associated with cells involved in human disease. Antigens associated with cells involved in human disease include cancer-associated tumor antigens and antigens associated with cells involved in autoimmune diseases. [0003] It was recognized in the design of these vaccines that a vaccine that produces the target antigen in the vaccinated individual is effective at inducing the cellular arm of the immune system. Specifically, live attenuated va...
