Embodiment Construction

[0032] Take the protoplast separation and fusion of Mexican wild species S. pinnatisectum (A) and potato cultivar (B) as an example, the specific steps of the technical solution of the present invention are as follows:

[0033] 1. Enzyme maceration: prepare pretreatment solution and enzymatic hydrolysis solution, for potato cultivar B, add maceration enzyme Macerozyme R-10 and cellulase Onozuka R-10 with a concentration ratio of 1:4 in the enzymolysis solution, The concentrations were 0.25% and 1%, respectively, and sterilized by a 0.2 μm filter sterilizer. Use the leaves of 24-40-day seedling-age potato wild species and the virus-free test-tube plantlets of cultivated varieties for tissue culture, cut the leaves into thin slices with a blade before enzymolysis, and place them in the pre-treatment solution for 1-2 hours. Then put the leaves into macerating enzymes and treat them for 16-30 hours under the condition of complete darkness at 20°C-30°C. Protoplasts were purified b...

Abstract

The invention provides a cultivating method of a new species of an anti-late blight and anti-beetle potato, relating to a cultivating method of a new species of the potato with a cell culture technology. The method comprises the following steps of: electrically fusing a protoplast (A) which has anti-late blight gene, anti-beetle gene and anti-oxidation gene, and is separated from the wild species of the potato, with a protoplast (B) of the potato culturing species with wide adaptability to form into a fusion cell with the protoplast A and the protoplast B; generating a callus by means of cell culture, and generating a hybrid test tube plantlet of a regenerated somatic cell by means of induction; and further culturing the tissue culture test tube plantlet, transplanting the cultured tissue culture test tube plantlet into a glasshouse or a land for growing field crops, and generating a test tube first generation somatic cell hybridized potato to obtain the anti-diseases and insect cell hybrid potato, wherein the somatic cell hybridized potato has the good agronomic trait of the potato cultivating species and the anti-late blight and anti-beetle trait. The cultivating method provides a new available gene resource which can be used for improving the quality of the potato and improving the antioxidant activity of the potato cultivating species with the wild species.

Description

technical field [0001] The invention relates to a method for cultivating new potato varieties using biological cell culture technology. technical background [0002] Potato late blight is the number one disease in potato production in the world, and the annual cost of controlling late blight is as high as 3 billion US dollars. The varieties cultivated by traditional breeding methods are all susceptible to late blight, and if not controlled, the yield loss can reach 100%. At present, high late blight resistance genes have been found in wild potato species, especially some wild species from Mexico, which can be used to improve the resistance of cultivated potatoes. In particular, a wild Mexican species called Solanum pinnatisectum has not only high late blight and beetle resistance genes, but also high antioxidant activity. It is an extremely precious and rare gene resource for disease resistance, insect resistance and antioxidant. However, due to its extremely unique genet...
