Problems solved by technology

The sensor has been greatly improved in terms of stability, sensitivity, selectivity, and reliability. By changing the nature and type of the receptor, it can be used for blood group analysis, detection of bacteria, viruses, DNA, drugs, antibodies, and electrolytes, etc., but Sensitivity and detection limit are far from meeting the needs of existing technology development

Embodiment Construction

[0027] The present invention will be further described in detail with reference to the accompanying drawings and embodiments.

[0028] The detection method provided by the invention is a method for preparing a biosensor to detect thrombin by combining an ion channel and an aptamer. The specific principles are as follows:

[0029] Such as figure 1 As shown, in the present invention, the ion channel 101 is prepared on the PET membrane 1, the pore size of the ion channel 101 is 150nm-200nm, and the inner wall of the ion channel 101 is modified with an aptamer 2 capable of specifically binding to the target object 4 . The DNA sequence of the adapter 2 is 5'-(NH 2 )-(CH 2 ) 6 -CCA TCT CCA CTT GGT TGG TGT GGTTGG-3'. The terminal modified amino group is designed to combine with the carboxyl group on the PET film 1. Then [Ru(NH 3 ) 6 ] 3+ (in the attached figure 1 The number in is 5 for illustration) modified on the phosphate backbone of DNA, and then sprayed gold on one sid...

Abstract

The invention discloses a method for detecting thrombin through a biosensor prepared based on an ionic channel and an aptamer, belonging to the field of biosensors. The method comprises the steps of: modifying a DNA (Deoxyribonucleic Acid) aptamer on an ionic channel, processing by using a P-ATP () mixture solution, and finally performing metal spraying to obtain a product serving as a work electrode, thereby obtaining the biosensor based on the ionic channel and the aptamer for detecting the thrombin. The biosensor disclosed by the invention has the advantages of simplicity, rapidness, high sensitivity and good selectivity, can be used to perform detection on actual samples, and has the minimal detection concentration reaching 1pM for the thrombin and a larger linear range. Within the concentration range from 3nM to 50nM, a linear relation exists between detected square wave volt-ampere peak current and the logarithm of concentration; the peak current value is stable, and the difference between the peak current value at the 10th circle and the peak current value at the 100th circle is within 3%; and the biosensor has very strong specificity, repeatability and practicability and has the recovery rate being 92-105% for the actual samples.

Description

technical field [0001] The invention belongs to the field of biosensors, and in particular relates to a method for preparing a biosensor based on an ion channel and an aptamer to detect thrombin. Background technique [0002] Thrombin is a multifunctional serine protease that plays an important role in a series of physiological and pathological processes in organisms. It not only participates in hemostasis and coagulation, inflammation, immune response, tissue repair and wound healing, but also activates the tumorigenic potential of normal cells and leads to the metastatic phenotype of malignant cells. With the development of modern society, people's material living standards continue to improve, but it is followed by the invasion of various diseases. And a large part of them are cardiovascular and brain diseases. Due to the limited detection technology, many curable diseases have lost the best treatment period, resulting in irreparable consequences. Recent research has sh...
