Problems solved by technology

The traditional hot water extraction method and acid-base extraction method require long extraction time, high extraction temperature, and high energy consumption, and long-term high temperature or acid-base medium may cause polysaccharides to degrade, making polysaccharide extraction difficult. To achieve the desired effect, these greatly limit the development and utilization of polysaccharides

Embodiment Construction

[0022] (1) Experimental design

[0023] 1. Hot water extraction method

[0024] With reference to Zhan Jianbo et al.’s report, the optimal conditions for extracting polysaccharides from Flammulina velutipes as raw materials using traditional hot water were slightly modified. The extraction temperature was 80°C, the extraction time was 4 hours, and when the ratio of water to material was 28ml / g, the yield of polysaccharides was 9.51. %.

[0025] 2. Ultrasonic extraction method

[0026] The ultrasonic power, ultrasonic time, and water-material ratio were selected as single-factor experiments, and the suitable ranges of the three variables were preliminarily determined through the single-factor experiments, and the three-factor and three-level Box-Behnken experimental design was carried out. The experimental levels and codes of each factor are listed in Table 1. , Box-Behnken experimental design and its response values are shown in Table 2.

Abstract

The invention relates to a method for extracting polysaccharides with cell immunocompetence from needle mushroom root waste materials and belongs to the field of biomedicine. The method comprises the steps of: drying and crushing needle mushroom roots to prepare ultrafine needle mushroom root powder, wherein the needle mushroom roots are the cut root parts as waste materials in a needle mushroom processing course; adding a proper amount of 85% ethanol to the crushed ultrafine powder to soak for 2 hours at room temperature; centrifuging to collect a precipitate; adding deionized water according to a ratio of water to material namely 28mL / g based on the raw material as a reference; carrying out ultrasonic treatment for 19.8 minutes at the power of 680W; centrifuging to collect a supernate; carrying out rotary evaporation on the supernate to obtain a proper amount of at room temperature; adding anhydrous ethanol the volume of which is 4 times that of a concentrated solution in the concentrated solution; precipitating for 12 hours at 4 DEG C; centrifuging to collect a precipitate; and placing the precipitate in an oven of 60 DEG C to bake so as to obtain crude needle mushroom root polysaccharide. An ultrasonic wave extraction technology is adopted, and the polysaccharide yield is 16.20%. Compared with the traditional hot water extraction, by using the method disclosed by the invention, the yield is improved by 70%, and the time is shortened by 92%.

Description

1. Technical field [0001] The invention relates to a method for extracting polysaccharides with cellular immune activity from waste materials of Flammulina velutipes mushrooms, which belongs to the field of biomedicine, and the product can be widely used in the fields of functional food, biomedicine and the like. Wherein the mushroom root of Flammulina velutipes is the root pedicle part that is removed as waste during the processing of Flammulina velutipes. 2. Background technology [0002] Flammulina velutipes (Flammulina velutipes), also known as Pleurotus velutipes and Shiitake mushrooms, belongs to the Phylum Fungi, Phytomycetes, Agaricaceae, Tricholomaceae, and Chrysostomyces. It contains high protein, carbohydrates and crude fiber, while the fat content is low. It also contains VB1, VB2, VC and other vitamins, and is rich in calcium, phosphorus, iron and other minerals. It is a rare high-nutrition product. [0003] As human beings gradually deepen their understanding ...
