Problems solved by technology

At present, green algae (such as Scenedesmus obliquus) are mainly used as the main food for artificial cultivation and toxicological evaluation of Daphnia magna, and it is impossible to accurately evaluate the true impact of insoluble solids such as transgenic rice on Daphnia magna

Embodiment Construction

[0016] Evaluating the biosafety of transgenic rice by feeding pure rice to Daphnia magna. The rice seeds selected for the experiment were "Bt Shanyou 63" and its parent seeds, and the foreign gene inserted was Cry1Ab / Cry1Ac fusion gene.

[0017] After long-term cultivation and domestication of Daphnia magna, young fleas of 6-24 hours with the same genetic background for more than three generations were selected as experimental fleas. The experimental fleas were cultured in a constant temperature culture room with a temperature of 22±1°C, a light intensity of 60 μmol m-2s-1, and a light-to-dark ratio of 16h:8h. Experimental culture solution formula: 294.0mg·L-1CaCl2; 123.2mg·L-1MgSO4; 64.8mg·L-1NaHCO3; 6.3mg·L-1KCL, the pH of the newly prepared culture solution is 7.8±0.2, and the hardness is 250±25mg·L -1 (calculated as CaCO3). Each treatment group was added 100ul of rice flour solution with different concentrations every day, (0.8g·L-1, 1.2g·L-1, 1.6g·L-1 rice flour, the dr...

Abstract

The invention relates to a breeding method for an aquatic organism daphnia magna, and belongs to the technical field of insect breeding. The method is used for overcoming the defect in a traditional daphnia magna toxicity experiment technology, and the invention builds a method for breeding daphnia magna by utilizing rice. The method comprises the following steps of: preparing a nutrient solution, preparing a rice flour solution, breeding and collecting young fleas. According to the daphnia magna breeding method disclosed by the invention, rice is used for breeding the daphnia magna, so that the defect that the daphnia magna in the conventional method selectively takes in natural goods such as green alga, but does not take in or take in a little rice is overcome. Moreover, an adverse factor that the experimental result is easily affected by uncertain nutritional ingredients of the green alga, and reliable experiment support is provided for evaluating the safety of transgenic rice.

Description

Technical field: [0001] The invention relates to a method for raising aquatic organisms Daphnia magna, belonging to the technical field of insect feeding Background technique: [0002] As one of the most important food crops in the world, rice provides food for nearly 3 billion people on the earth, but pests and diseases seriously affect the yield and quality of rice. Introducing exogenous genes such as insect resistance and disease resistance into the rice genome through transgenic technology, and cultivating new insect-resistant and disease-resistant transgenic rice varieties has become a new method and new way to prevent and control rice diseases and insect pests and increase rice yield. With the large-scale promotion of genetically modified crops, people pay more and more attention to its safety. Due to the important position and influence of my country's rice production and consumption in the world, the Chinese government is very cautious and serious about the commerci...
