[0037] The protocol for this study and the acquisition of tissue samples were approved by the Biomedical Research Ethics Committee of the Second Military Medical University, Shanghai, China. Glioma tissue samples were derived from previously collected patient tissue samples from patients with glioma who underwent surgery in the Neurosurgery Department of Shanghai Changzheng Hospital between January 1999 and December 2008. Normal brain tissue samples were obtained from patients with severe brain trauma in which part of the normal brain tissue had to be removed to relieve intracranial pressure. Abstract

The invention relates to a method for detecting expression of bone sialoprotein (BSP) in brain glioma by utilizing a high-throughput tissue chip. The method comprises the following steps of: (1) performing total RNA (ribonucleic acid) separation on brain tissues by using a Trizol reagent; (2) performing inverse transcription on the total obtained RNA to obtain cDNA (complementary deoxyribonucleic acid); (3) mixing the cDNA and SYBR fluorescent dye and carrying out amplification; (4) standardizing expression of IBSP through GAPD, and carrying out a comparison threshold cycle method by using an equation; and (5) establishing the tissue chip, performing immumohistochemical staining, performing statistic analysis on the immumohistochemical result, and detecting expression of BSP in the brain glioma. According to the method, the sensitivity of detecting the expression of BSP is greatly improved, and enough information can be acquired in few specimens; and moreover, the method has an important medical value for detecting the expression of BSP.
