Problems solved by technology

However, these methods have more or less shortcomings such as long detection period, complicated procedures, and a large number of reagents required. If the sample volume is large, these technologies are difficult to meet the needs of rapid counting of Salmonella

Embodiment 1

[0037] 1) Obtain multiple Salmonella solutions with different known concentrations:

[0038] Take the first concentration of Salmonella bacteria solution and dilute it with sterile saline for 10 3 、10 4 、10 5 、10 6 、10 7 、10 8 Times, each take 200 μL of the dilution of Salmonella into 12 Petri dishes (parallel samples for each dilution). Spread the dilutions in each culture dish on LB plates, incubate in an incubator at 35-37°C for 24 hours, count the number of suspicious colonies on each plate, pick suspicious colonies for biochemical and serological identification verification, A solution of Salmonella of known concentration is obtained. The second concentration was cultured and analyzed in the same way, and a series (7) of Salmonella solutions with different known concentrations were obtained by analogy.

Abstract

The invention discloses a method for rapidly detecting the concentration of salmonellae in water. The method comprises the following steps of: (1) obtaining a plurality of parts of salmonella solutions with different known concentrations; (2) rapidly detecting the concentration of the salmonellae by using an electrochemical chronoamperometry through a CHI660D electrochemical workstation; determining that a compliance relation between the salmonellae and boron-doped diamond film electrode responding current is a linear relation, wherein parameters of the CHI660D electrochemical workstation are that: a boron-doped diamond film electrode is used as a working electrode, the detection potential is 0.9-1.15V and an electrolytic tank is filled with a PBS (Phosphate Buffer Solution) with the pH (Potential of Hydrogen) being 7.2-7.6; and (3) carrying out sample introduction on a water body to be detected and recording a current value of responding electrophoresis of the boron-doped diamond film electrode; and detecting the salmonella concentration corresponding to the current value by utilizing an obtained responding current-salmonella concentration curve and combining the current value of the sample. The method disclosed by the invention is simple to operate; the detection time is only 1 hour; and the detection limit can reach 1000cfu / mL.

Description

technical field [0001] The invention relates to a method for determining the content of microorganisms, in particular to a method for rapidly detecting the concentration of Salmonella in a water body. Background technique [0002] Salmonella belongs to the Enterobacteriaceae family, which includes those bacteria that cause food poisoning, gastroenteritis, typhoid fever, and paratyphoid fever. In addition to infecting humans, salmonellosis can also infect many animals including mammals, birds, reptiles, fish, amphibians and insects. Human and animal infection can be asymptomatic carrier state, or manifest as fatal disease with clinical symptoms, which may aggravate morbidity or mortality, or reduce animal reproductive productivity. [0003] At present, the concentration of Salmonella in water is generally detected by traditional routine detection methods, mainly through bacterial enrichment, separation, and biochemical identification, such as plate counting, etc. These metho...
