Problems solved by technology

[0005] At present, the research and development of ODP-related products has gradually begun at home and abroad, but the food research and development is mainly based on fresh cactus fruit juice, and there are few researches on dosage forms, and the research on ODP microspheres has not been reported yet.

Embodiment 1

[0024] First, dissolve 2 g of gelatin in 20 mL of water, then add 200 mg of wild cactus polysaccharide, and mix thoroughly at 60°C to make a water phase solution (W); take 60 mL of liquid paraffin, add Tween-80 with a volume concentration of 1% The emulsifier is formed as the oil phase continuous phase (O); the gelatin polysaccharide water phase is slowly added to liquid paraffin at a ratio of W / O of 1:3 and fully stirred for 35 min on a magnetic stirrer (stirring speed 1000 r / min). Form W / O emulsion; after fully emulsified, cool rapidly below 5°C and keep stirring for 35min, add 0.8mL of 25% glutaraldehyde to solidify for 2h. Finally, centrifuge, take the precipitate and wash with isopropanol or ethanol to remove residual oil and glutaraldehyde, and freeze-dry to obtain microspheres. The drug loading and encapsulation efficiency were determined to be 7.47% and 91.17%, respectively.

Embodiment 2

[0026] The steps are basically the same as in Example 1, except that the W / O ratio is 1:4, the stirring time is 40 min, and the stirring speed is 1000 r / min.

[0027] The prepared gelatin microspheres had a drug loading capacity and encapsulation efficiency of 7.39% and 90.62% respectively.

Embodiment 3

[0029] The steps are basically the same as in Example 1, except that: when preparing the water phase, the amount of wild cactus polysaccharide added is 100 mg, and the W / O is 1:3 and stirred for 45 min at a stirring speed of 1000 r / min. The prepared gelatin microspheres had a drug loading capacity and encapsulation efficiency of 4.64% and 92.41%, respectively.

Abstract

The invention relates to high-encapsulation-rate wild cactus polysaccharide gelatin microspheres and a preparation method thereof. The preparation method comprises the steps that: a small amount of gelatin is dissolved into a proper amount of water; a wild cactus polysaccharide solution is added; the materials are sufficiently mixed and pre-emulsified, such that a water-phase solution is prepared; an oil-phase continuous phase is liquid paraffin, and a small amount of tween-80 is added as an emulsifier; on a basis that original stable condition is maintained, the gelatin polysaccharide water phase is slowly added into liquid paraffin, and the mixture is well mixed; after sufficient emulsification, the mixture is rapidly cooled to below 5 DEG C while stirring is continued; glutaraldehyde is added, and curing is carried out for 2h; centrifugation is carried out and precipitate is fetched; residual oil and glutaraldehyde are removed by washing by using isopropanol or ethanol; and lyophilization is carried out, such that microspheres are obtained. With the method, drug load and encapsulation rate of wild cactus polysaccharide microspheres respectively reaches 7.47% and 91.38%. The method has the advantages of simple process, low cost, and high repeatability. The prepared wild cactus polysaccharide gelatin microspheres has round appearance, uniform particle size, good dispersion, high encapsulation rate, good biocompatibility, and good biodegradability.

Description

technical field [0001] The invention relates to a polysaccharide gelatin microsphere and a preparation method thereof, in particular to a gelatin microsphere prepared by using wild cactus polysaccharide and a preparation method capable of improving its encapsulation rate, and belongs to the technical field of biomedicine preparation. Background technique [0002] Our wild cactus ( Opuntiadillenii Haw. ) is widely distributed and mainly grows in mountains, islands and desert areas. It was first recorded in "Compendium of Materia Medica" (the ninth volume of grass in the twentieth volume of "Tu Jing" in Song Dynasty), and later in "Compendium of Materia Medica Supplements". It has been confirmed in its various effects such as promoting qi and blood circulation, clearing heat and detoxification, reducing swelling and relieving pain, invigorating the spleen and stopping diarrhea, calming the nerves and diuresis. Modern biological research has found that one of the most importa...
