Problems solved by technology

[0013] In our previous work, we have studied HGF by SDS-page, Western blot, ELISA and SPR, and showed that HGF protein that does not bind to proteoaminoglycans (HSPG, heparan sulfate) or dextran sulfate (endo source or recombinant) had no biological effect when used in vivo (hair growing mice) or in vitro bioactive methods (CCL-53.1 cells) by our research group

Embodiment

[0054] A test strip with two surfaces for detection of affinity for HSPG and pH was developed according to the present invention. In this example, the strip test has been evaluated to identify infectious gastroenteritis from other causes of diarrhea. The correlation of fecal pH with survival and the correlation of test results with fecal calprotectin was evaluated in a cohort of patients with SIRS in the detection of several chronic inflammatory diseases. The sensitivity of conventional microbiological methods to identify infectious gastroenteritis was evaluated retrospectively in patient material admitted and discharged to our ward in 2011.

[0055] research group

[0056] Stool samples from all patients with bowel disorders in Östergötland, Sweden, retrieved from health institutions were included in the study. There are no exclusion criteria. Samples are immediately numbered and unidentified. Patients (n = 140) retrieved from a health care center between March 3 and Augu...

Abstract

The present invention relates to a method for determining the presence, absence or amount of biologically active HGF in a sample, comprising the steps (i) bringing the sample in contact with a first porous solid phase comprising a HGF binding component of the extracellular matrix or cell membrane, and an indicator composition comprising bromothymol blue and a quaternary ammonium compound; and (ii) correlating the colour of the porous solid phase with the presence, absence or amount of biologically active HGF in the sample. It further relates to a device comprising a first porous solid phase comprising at least one HGF-binding component of the extracellular matrix or cell membrane, and an indicator composition comprising bromothymol blue and a quaternary ammonium compound.

Description

field of invention [0001] The present invention belongs to the technical field of products for detecting the presence of growth factors such as hepatocyte growth factor (HGF) in biological samples. The invention also relates to methods of carrying out such methods. Background of the invention [0002] Hepatocyte growth factor (HGF) is a unique growth factor that is not related to other known polypeptide mitogens. It is a protein expressed in mesenchymal cells such as lung macrophages and fibroblasts, Kupffer cells in the liver, and leukocytes. HGF is secreted in response to cellular injury and appears to be important for regeneration of certain organs and wound healing. It is a heterodimer with disulfide-bonded heavy and light chains of approximately 60 and 30 kDa, respectively, which is first synthesized as an inactive precursor. In damaged organs, the precursor is cleaved into active protein by specific activators. HGF acts in a paracrine form (ie it affects neighborin...
