[0029] (1) Synthesis of PGS-P

[0030] ① Synthesis of PGS

[0031] PGS synthesis reference (Wang YD, Ameer GA, Sheppard BJ, Langer R.A toughbiodegradable elastomer. Nature biotechnology.2002; 20:602-6.), and its optimization. After recrystallization and purification, 20.2574 g (0.1 mol) of sebacic acid (J&K, 98%) and 9.2037 g (0.1 mol) of glycerin (Sigma Aldrich, ≥99.5%) were added into a three-necked flask, melted at 135 ° C, and passed through N 2 Stir for 24h, then evacuate (4mbar) at 135°C for 48h, cool down to room temperature to give a pale yellow waxy solid.

[0032] ② Synthesis of PGS-P

[0033] Weigh 0.6865g (2.6609mmol, the formula weight of a chain segment containing a hydroxyl group as the molecular weight) PGS is added to the 100ml flask that has been dewatered and deoxygenated in advance, and then 0.1631g (1.0637mmol) POCl3 has been weighed and added to the dehydrated and deoxygenated flask. Into the 10ml reaction tube, add 20ml and 7ml of anhydrous tetrahydro...

Abstract

The invention relates to a degradable phosphorylated material, a preparation method thereof, and applications of the degradable phosphorylated material in osteogenesis. The degradable phosphorylated material is poly(glycerol sebacate)phosphate PGS-P with phosphorus content larger than 0% and lower than 9.14%. The preparation method comprises following steps: (1) synthesis of PGS, and (2) synthesis of PGS-P. The degradable phosphorylated material can be used for preparation of tissue engineering scaffolds. Synthesis of the degradable phosphorylated material is simple; structure of the degradable phosphorylated material is controllable; phosphorylation degree of the degradable phosphorylated material can be adjusted effectively; the degradable phosphorylated material possesses excellent osteogenic activity, and the osteogenic activity can be adjusted via adjusting of phosphorylation degree; a novel material is provided for biomedicine application such as osteanagenesis, and design thought of the degradable phosphorylated material can be applied to design of other similar phosphorylated materials and osteogenesis active materials; the preparation method is simple; cost is low; and application prospect is promising.

Description

technical field [0001] The invention belongs to the field of bone tissue engineering scaffold materials, and in particular relates to a degradable phosphorylated material, a preparation method thereof and an osteogenesis application. Background technique [0002] As one of the essential elements for life, phosphorus is one of the constituent elements of bones, teeth, and genetic material nucleic acids. Phosphorylated materials have a wide range of applications in the field of biomaterials, especially in the field of bone defect repair. Bone defect is one of the most common and thorny clinical problems, which can be caused by traumatic fractures, inflammation, tumors, congenital abnormalities, etc. In the United States alone, 6.2 million fractures occur a year, of which 5-10% cannot be effectively healed [Kretlow JD, Mikos AG. Review: Mineralization of synthetic polymer scaffolds for bone tissue engineering. Tissue Eng. 2007; 13:927-38 .], regenerative medicine is a potentia...
