Embodiment 1

[0038] A kind of embodiment of the detection method of human epidermal growth factor receptor-2 concentration of the present invention, its schematic diagram is as figure 1 shown. The detection method specifically includes the following steps:

[0039] (1) Pretreatment of gold electrodes: First, polish multiple gold electrodes with polishing powder with a particle size of 0.3 μm for 5 minutes, and then perform secondary polishing with polishing powder with a particle size of 0.05 μm. Wash with water (water after two distillations), then ultrasonically use absolute ethanol for 2 minutes, and then use secondary water for 2 minutes to confirm that there is no residual polishing powder on the surface of the gold electrode, then take out the gold electrode and rinse it with secondary water, and then use nitrogen gas Blow dry and get more pre-treated gold electrodes.

[0040] (2) Polypeptide self-assembly: Insert multiple pretreated gold electrodes upside down into a centrifuge t...

Embodiment 2

[0048] A kind of embodiment of the detection method of human epidermal growth factor receptor-2 concentration of the present invention, its schematic diagram is as figure 1 shown. The detection method specifically includes the following steps:

[0049] (1) Gold electrode pretreatment: Firstly, multiple gold electrodes are polished with polishing powder with a particle size of 0.3 μm for 6 minutes, and then polished with a polishing powder with a particle size of 0.05 μm for a second time. Wash with water (water after two distillations), then ultrasonically use absolute ethanol for 3 minutes, and then use secondary water for 3 minutes to confirm that there is no residual polishing powder on the surface of the gold electrode, then take out the gold electrode and rinse it with secondary water, and then use nitrogen gas Blow dry and get more pre-treated gold electrodes.

Abstract

The invention discloses a method for detecting the concentration of human epidermal growth factor receptor-2 (HER-2). The method comprises the following steps: grinding, cleaning and drying gold electrodes; inserting the gold electrodes into a polypeptide solution for carrying out self-assembly, and then closing the gold electrodes with mercapto hexanol; respectively dripping HER-2 with different concentrations onto the surfaces of the gold electrodes for carrying out reactions, and then enabling the gold electrodes to react with an aptamer; dripping molybdate onto the surfaces of the gold electrodes for carrying out reactions to obtain a plurality of to-be-detected gold electrodes; respectively measuring the gold electrodes by using a square wave voltammetry to obtain a plurality of square wave voltammetric curves, and making a standard curve according to peak currents in all the curves and the corresponding concentrations of the HER-2; dripping the HER-2 to be detected onto the surface of the gold electrode assembled with polypeptide for carrying out a reaction, then enabling the gold electrode to react with the aptamer, dripping molybdate onto the surface of the gold electrode, measuring by using the square wave voltammetry to obtain a square wave voltammetric curve, and contrasting the peak current with the standard curve to obtain the concentration of the HER-2. The detection method is convenient and fast, good in selectivity, high in sensitivity and wide in detection range.

Description

technical field [0001] The invention relates to the technical field of biosensing, in particular to a method for detecting the concentration of human epidermal growth factor receptor-2. Background technique [0002] Human epidermal growth factor receptor-2 (human epidermal growth factor receptor-2; HER-2) is a receptor tyrosine kinase that is highly expressed in a variety of human cancers. It is a special marker protein of breast cancer, closely related to the metastasis of breast cancer, and can effectively judge the prognosis of breast cancer. [0003] Receptor tyrosine kinases of the epidermal growth factor-related protein family play an important role in epithelial growth as major therapeutic targets in several cancers. Overexpression and mutation of EebB (HER) family members can lead to a large number of malignant tumors. Traditional methods for measuring HER-2 include surface plasmon resonance, immunohistochemistry, and enzyme-linked immunosorbent assay. Although the...
