The method comprises the steps: (1) making a solution containing polyfructose contact with amylase, to obtain a first solution; (2) making two equivalent parts of the first solution respectively contact with a dextranase/fructozyme composite enzyme solution and an acetate buffer solution, to obtain a second solution and a third solution; (3) carrying out cooling treatment on the second solution and the third solution; (4) diluting the second solution and the third solution after the cooling treatment in the step (3), to obtain a diluted solution A and a diluted solution B; and (5) testing the fructose concentration in the diluted solution A, and the sucrose concentration and the fructose concentration in the diluted solution B by ion chromatography, and calculating to obtain the content of polyfructose.
