Embodiment 1

[0015] (1) Bud induction culture: Take the stem section of the thousand-year-old healthy belt bud as an explant, cut off the leaves, and cut it into a 5.8-inch section with 6 axillary buds. Soak it in washing powder solution for 8 minutes, and then gently brush it with a soft brush. Lightly brush the surface of the explant to remove the dust and part of the bacteria on the surface, then rinse it with tap water for 3 hours, then place it in an ultra-clean workbench, first disinfect it with 75% ethanol for 8 seconds, then wash it with sterile water for 6 times, and then use 0.1% L Mercury solution was sterilized for 13 minutes, rinsed with sterile water for 7 times, and then wiped dry the water droplets on the surface with sterile filter paper, and then inoculated into bud induction medium for bud induction. Illuminated for 13 hours, and the light intensity was cultivated for 33 days under the condition of 1500lx, and the bud induction rate and growth situation were counted, and ...

Embodiment 2

[0020] (1) Bud induction culture: Take the stem section of the thousand-year-old healthy belt bud as an explant, cut off the leaves, cut into 4.9 long segments with 4 axillary buds, soak them in washing powder solution for 9 minutes, and then gently brush them with a soft brush. Lightly scrub the surface of the explant to remove the dust and part of the bacteria on the surface, then rinse it with tap water for 3 hours, then place it in an ultra-clean workbench, first disinfect it with 75% ethanol for 14 seconds, then wash it with sterile water for 4 times, and then use 0.1% L Sterilize with mercury solution for 14 minutes, rinse with sterile water for 3 times, then dry the water drops on the surface with sterile filter paper, and then inoculate into bud induction medium for bud induction. Illuminated for 9 hours, and the light intensity was cultivated for 28 days under the condition of 1300lx, and the bud induction rate and growth situation were counted, and the bud induction r...

Abstract

The invention discloses an obscured homalomena rhizome tissue culture method. An obscured homalomena rhizome is an Araceae plant and is the dry rhizome of homalomena occulta (Lour.) Schott. The methodcomprises the steps that rhizomes are harvested in Spring and Autumn, leaves and seedlings are removed, soil is cleaned away, the processed rhizomes are dried, or the rhizomes are dried after the skin is scraped off. The rhizomes are shaped like cylinders or are slightly flat and bend slightly. The surface is rufous or yellowish-brown; the rhizomes are rough and have multiple twisted longitudinalgrooves and yellow-white fiber bundles; the rhizomes are crisp and likely to be broken, the cross section is rufous, seems like resin and has many fiber bundles which are exposed and circular oil spots with luster. The smell is fragrant, and the taste is acrid and slightly bitter. The best rhizomes are hard, rufous and rich in fragrance. The rhizomes contain volatile oil including pinene, linalool and perilla aldehyde. Accordingly, stems, with buds, of the obscured homalomena rhizome serve as explant, regeneration in vitro of the obscured homalomena rhizome is achieved through the processes such as bud induction, bud-propagation culture, rooting culture, seedling exercising and transplanting, and technical support is provided for a large amount of breeding, rapid propagation and new species popularization. The technical method is simple, operation is easy, seedlings are thick and strong, growth is rapid, and the yield is high.

Description

technical field [0001] The invention belongs to plants, and in particular relates to a method for culturing healthy tissue of a thousand years. Background technique [0002] Qiannianjian is an araceae plant, the dry rhizome of Qiannianjian. It is mainly produced in southern Guangxi and Honghe, Xishuangbanna and other places in Yunnan. The rhizomes are excavated in spring and autumn, the leaves and seedlings are removed, the soil is washed, and dried in the sun, or scraped off the outer skin and then dried in the sun. Rhizome cylindrical or slightly flattened, slightly curved. It is 15~40cm long and 0.8~2cm in diameter. The surface is reddish brown or yellowish brown. Rough, with many twisted longitudinal grooves and yellow-white fiber bundles. Brittle, easy to break, reddish-brown cross-section, resinous, with many fiber bundles exposed, and round shiny oil spots. Gas aroma, pungent, slightly bitter. Those with hard quality, reddish-brown color and strong aroma are pr...
