Abstract

The invention relates to the field of plant tissue culture, provides a tissue culture method for avoiding crown inclination of cunninghamia lanceolata test-tube plantlets, and solves the defect of crown inclination of test-tube plantlets cultured in the prior art. The method comprises the following steps: (1) obtaining explants; (2) performing proliferation culture; (3) performing seedling strengthening culture; (4) performing rooting culture; (5) hardening seedlings: hardening the seedlings under the condition of a 60% shading degree, when root systems change colors, keeping the humidity of agreenhouse to be 70% or more, opening a cover, and hardening the seedlings for 3 days; and (6) transplanting the test-tube plantlets: when transplantation is performed, rinsing the rooting seedlingswith fresh water, draining off water, soaking the treated seedlings into a 0.2% potassium permanganate aqueous solution for 5min, lifting the seedlings, rinsing the seedlings with fresh water for 3 times, putting the rinsed seedlings into seedling raising mixed matrix nutrient bags, performing culture, and when the heights of the seedlings cultured in containers reach 30cm, enabling the seedlingsto leave a nursery for afforestation.
