The preparation method comprises the following steps: (1) establishment of a rutin standard curve; (2) extraction of astragalus membranaceus flavonoids; and (3) calculation of astragalus membranaceus flavonoid concentration. The provided duck natural immune enhancer based on the traditional Chinese medicine extract can significantly increase levels of duck serum cytokines of IL-1-beta, IFN-alpha and IFN-beta, up-regulates tissue expression levels of natural immune-related genes RIG-I, TLR3, TLR7, IFN-alpha, IFN-beta, IL-1-beta and TNF-alpha, inflammation-related genes COX2 and iNOS, and cell apoptosis-related gene Caspase3 in duck immune organs of liver, spleen, thymus and bursa of fabricius, and enhances duck natural immune response and disease resistance performance.
