Embodiment 1

[0033] Example 1 Preparation of spindle-shaped BiVO 4 ,Proceed as follows:

[0034] 1 mmol Bi(NO 3 ) 3 ·5H 2 O was ultrasonically dispersed in 50 mL of ethylene glycol, and then 1 g of polyvinylpyrrolidone (PVP; MW ~ 40K) was slowly added to the above solution under magnetic stirring, and ultrasonically dispersed for 20 min, labeled as solution A; 1 mmol NH 4 VO 3 Disperse in 30 mL of ultrapure water and disperse under ultrasonic for 10 minutes, and mark it as solution B; slowly add solution B to solution A under magnetic stirring, and the color changes from transparent to orange; the final mixture is transferred to 100 mL poly The reaction was carried out in a vinyl fluoride reactor at 180 °C for 10 h, and the resulting mixture was centrifuged and washed three times under ultrapure water and absolute ethanol, and then dried at 50 °C.

Embodiment 2

[0035] Example 2 Preparation of spindle-shaped BiVO 4 ,Proceed as follows:

[0036] 1.5 mmol Bi(NO 3 ) 3 ·5H 2 O was ultrasonically dispersed in 50 mL of ethylene glycol, and then 2 g of polyvinylpyrrolidone (PVP; MW ~ 40K) was slowly added to the above solution under magnetic stirring, and ultrasonically dispersed for 20 min, labeled as solution A; 1.5 mmol NH 4 VO 3 Disperse in 30 mL of ultrapure water and disperse under ultrasonic for 10 minutes, and mark it as solution B; slowly add solution B to solution A under magnetic stirring, and the color changes from transparent to orange; the final mixture is transferred to 100 mL poly The reaction was carried out in a vinyl fluoride reactor at 180 °C for 10 h, and the resulting mixture was centrifuged and washed three times in ultrapure water and absolute ethanol, and then dried at 50 °C.

Embodiment 3

[0037] Example 3 Preparation of spindle-shaped BiVO 4 ,Proceed as follows:

[0038] 2 mmol Bi(NO 3 ) 3 ·5H 2 O was ultrasonically dispersed in 50 mL of ethylene glycol, and then 3 g of polyvinylpyrrolidone (PVP; MW ~ 40K) was slowly added to the above solution under magnetic stirring, and ultrasonically dispersed for 20 min, labeled as solution A; 2 mmol NH 4 VO 3 Disperse in 30 mL of ultrapure water and disperse under ultrasonic for 10 minutes, and mark it as solution B; slowly add solution B to solution A under magnetic stirring, and the color changes from transparent to orange; the final mixture is transferred to 100 mL poly The reaction was carried out in a vinyl fluoride reactor at 180 °C for 10 h, and the resulting mixture was centrifuged and washed three times in ultrapure water and absolute ethanol, and then dried at 50 °C.

Abstract

A photoelectrochemical immunosensor for detecting a dairy product allergen beta-lactoglobulin (beta-LG) is constructed, an Ag2S sensitized spindle-shaped BiVO4/BiOBr heterojunction is fixed on the surface of an ITO (indium tin oxide) electrode by using an antibody for specifically recognizing allergen beta-LG to be used as an excellent photoactive substrate, so that the photocurrent response and the sensitivity are effectively improved. Thioglycollic acid TGA is used as a connecting agent to fix the beta-LG antibody, photocurrent signals of antigens with different concentrations are detected through specific binding between the antigens and the antibody, and the quantitative test of the beta-LG is completed. Under the optimal experiment conditions, the PEC immunosensor is good in stability, wide in linear range, high in sensitivity and good in reproducibility, and can be used for detecting actual samples. The label-free immunosensor has very strong capability of specially identifying beta-LG, is not influenced by other proteins in milk, and does not need pretreatment. Meanwhile, the developed immunosensor provides a promising PEC detection platform and a reference thought for detection of other proteins in milk.

Description

technical field [0001] The invention relates to the technical fields of bioanalytical chemistry, nanometer materials, immunoassay and photoelectrochemical biosensing, and provides a preparation method and application of a photoelectrochemical biosensor for detecting β-lactoglobulin. Spindle-shaped BiVO synthesized by hydrothermal method 4 / BiOBr, Ag 2 S is used as a sensitizer, the antibody capture material is fixed on the surface of the electrode, and a preparation method and application of a label-free photoelectrochemical biosensor for detecting β-lactoglobulin are provided. Background technique [0002] Milk allergy (CMA) is one of the most important allergic reactions, and milk is also one of the most common allergens in food, characterized by a strong IgE reaction to milk protein. The prevalence of CMA in children under 2 years old is about 2.5%. CMA is a serious problem for children because cow's milk is an important source of protein for them, and strict avoidance...
