The method comprises the following steps:firstly, screening thiobacillus thiooxidans and thiobacillus ferrooxidans in sludge by using a thiobacillus thiooxidans culture medium and a thiobacillus ferrooxidans culture medium which are self-prepared; secondly, respectively carrying out amplification culture on the strain obtained by screening for three times by using the prepared culture medium according to different inoculation proportions; and finally, inoculating the bacteria subjected to amplification culture into required sludge for sludge domestication, and carrying out sludge domestication culture for three times to obtain the strain required by bioleaching. Compared with a traditional method, strain counting is not needed, the operation is easy and convenient, and the strain obtaining time is shorter.
