Embodiment 1

[0029] Example 1. A metagenomics-based approach for indigenous bacteria to remediate oil-contaminated soils as figure 1 As shown, proceed according to the following process steps:

[0030] (1) Screening, enrichment and cultivation of petroleum degrading bacteria:

[0031] (1.1) Culture medium preparation is the basic part of the present invention. Improved Basal Salt Medium (MSM): During the preparation process, it is strictly based on the MSM standard ratio, (NH 4 ) 2 SO 4 2.0g / L; MgSO 4 ·7H 2 O 0.2g / L; CaCl 2 2H 2 O 0.01g / L; FeSO 4 ·7H 2 O 0.001g / L; Na 2 HPO 4 12H 2 O 1.5g / L; KH 2 PO 4 1.5g / L; distilled water 1000ml; modified medium needs to add glucose 0.5g / L, tryptone 0.5g / L. Sterilize at 121°C for 20-40 minutes.

[0032] (1.2) Sample collection. Collect 10 contaminated soil samples near the oil depot (E121.5544, N41.9095) and 0-60cm from the ground, and collect at multiple points on the same plane. Immediately after the sample is collected, it is tran...

Embodiment 2

[0097] Example 2. It is an application example of a metagenomics-based method for remediating oil-contaminated soil by indigenous bacteria. Proceed according to the following process steps:

[0098] (1) The preparation of culture medium for mixed bacteria is the basic part of the present invention. Improved Basal Salt Medium (MSM): During the preparation process, it is strictly based on the MSM standard ratio, (NH 4 ) 2 SO 42.0g / L; MgSO 4 ·7H 2 O 0.2g / L; CaCl 2 2H 2 O0.01g / L; FeSO 4 ·7H 2 O 0.001g / L; Na 2 HPO 4 12H 2 O 1.5g / L; KH 2 PO 4 1.5g / L; distilled water 1000ml; modified medium needs to add glucose 0.5g / L, tryptone 0.5g / L. Sterilize at 121°C for 20-40 minutes.

[0099] (2) Sample collection. Ten contaminated soil samples near the original site of the Dalian Chemical Plant (E121.6051, N38.9579) and 60-200 cm above the ground were collected at multiple points on the same plane. Immediately after the sample is collected, it is transported to a refrigerato...

Abstract

The invention discloses a method for repairing petroleum-polluted soil by indigenous bacteria based on metagenomics, which comprises the following steps: (1) screening, enriching and culturing petroleum degrading bacteria by using benzene as an induction and screening condition; (2) carrying out diversity analysis on mixed culture bacteria; (3) determining the petroleum pollutant degradation capability of the mixed bacteria; (4) determining the degradation effect of the mixed bacteria on the soil petroleum pollutants and constructing an optimal degradation system; (5) performing soil metagenomics sequencing and data analysis; (6) carrying out soil metagenomics analysis on abundance inter-group differences of key degrading enzymes; and (7) a hydrogen donor degradation system construction method. Petroleum hydrocarbon degradation is realized through in-situ indigenous bacteria under normal-temperature and low-temperature conditions, namely benzene-induced mixed bacteria optimized degradation and hydrogen donor degradation. Hydrogen donor degradation can be continuously carried out, and the micro-ecological environment of the soil is slowly recovered; and no secondary pollution is caused in the soil ecological restoration process.

Description

technical field [0001] The invention belongs to the field of microbiological technical treatment and repair of environmental pollutants, and in particular relates to a method for remediating oil-contaminated soil by indigenous bacteria based on metagenomics. The main microbial group of the present invention belongs to bacteria, and is facultative trophic bacteria, which can utilize ready-made organic matter and petroleum hydrocarbon organic matter, and can tolerate the stress of toxic substances such as petroleum hydrocarbons. Background technique [0002] Petroleum is one of the most important fossil energy in modern society. Due to the rapid development of modern global industrialization, the demand for petroleum, which is called "industrial blood" by human beings, has increased by leaps and bounds. With the extraction, transportation, refining and utilization, a large amount of oil is leaked into the environment, causing economic losses and waste of resources. In nature,...
