Embodiment 1

[0025] (1) use the ethanol reflux extraction method to carry out the experiment, adopt L 9 (3 4 ) Orthogonal design experiment optimized the conditions such as ethanol concentration, solvent amount, extraction time and extraction temperature. Taking lutein as a contrast, the content of total flavonoids in the extract was determined by ultraviolet spectrophotometry, and used as a basis indicators for comparative analysis. The results are shown in Table 1, Table 2, Table 3, and Table 4.

[0031]

[0032] Note: F 0.1 (2, 2) = 19.00, F 0.05 (2, 2) = 9.0

[0033] The intuitive analysis and variance analysis of the above orthogonal test results show that the order of factors affecting the extraction effect of total flavonoids of P. chinensis medicinal...

Embodiment 2

[0038] Carry out following experiment with the Pseudomonas extract that obtains with aforementioned preferred condition extraction:

[0039] (1) Screening experiment of macroporous resin

[0040] Resin screening results are shown in Table 5. figure 1 and figure 2 .

[0041] Table 5 Static adsorption and desorption results of different macroporous adsorption resins on water yellow skin flavonoids

[0042]

[0043] D-101, AB8, and X-5 resins are all products of Anhui Sanxing Resin Technology Co., Ltd. The above results show that D101 and AB-8 resins are better than X-5 resins.

[0044] The adsorption capacity of D101 and AB-8 was investigated, with 0.6071mg.ml -1 The concentration of the sample was loaded at a flow rate of 2ml.min, and the effluent received 1 tube for every 5ml and was continuously taken. Carry out ultraviolet detection on the effluent, draw the leakage curve with the resin volume multiple of the effluent as the abscissa, and the total flavonoid concent...

Embodiment 3

[0068] Example 3 Pilot Scale-Up Test

[0069] Take 10 kg of the Chinese herbal medicine Pseudomonas, reflux and extract with 60% ethanol for 3 times, each time with 150 L of ethanol, extract for 0.5 h, combine the extracts for 3 times, filter, recover the ethanol, leave it overnight, and take the supernatant for later use.

[0070] Take the pharmaceutical grade D101 macroporous resin, soak it with an appropriate amount of ethanol, put it on the column by wet method, and treat it for later use.

[0071] Add the above-mentioned extracted medicinal solution to the resin column, the sample loading flow rate is 2ml.min-1, pH=3, after loading the sample, wash it with purified water at the same flow rate until the molish reaction is negative, and then elute with 3 times the column volume of 20% ethanol, Finally, it was eluted with 70% ethanol, and the end point of elution was determined by TLC method. The eluate was collected, concentrated with recovered ethanol, and dried in vacuum ...

Abstract

The invention discloses an extracting and purifying process of pongamia pinnata flavonoids. The extracting steps of the process are as follows: step one, taking roots, stems and coarse leaf powders of pongamia pinnata, adding 50-70% of ethanol solution according to the proportion (mass per unit volume) being 8-15 times of medicinal materials, refluxing and extracting the mixture under 60-80 EDG C by three times for 0.5-2.0h each time, combining and filtering an extracting solution, and decompressing and recycling ethanol so as to obtain a pongamia pinnata flabonoid extracting solution; step two, standing the pongamia pinnata flabonoid extract solution obtained from the step one, taking supernatant and utilizing macroporous absorption resin for purifying the pongamia pinnata flabonoid, and eluting by ethanol; and step three, collecting ethanol eluent obtained from the step two, decompressing and recycling the ethanol for concentration, conducting vacuum drying under the temperature of 50-65 DEG C, and finally obtaining the purified object of an extract. In the process, the mass of total flavone accounts for 50%-70% in the extract of pongamia pinnata, and the transferring rate of the total flavone in the pongamia pinnata is 75-80%. Compared with the prior art, the extracting and purifying process has the advantages that the process is simplified, the loss of active ingredients is reduced, the safety is higher, and the like.

Description

technical field [0001] The invention relates to a method for extracting and purifying the effective parts of medicinal plants, in particular to a method for extracting and purifying the medicinal material pumella and its extract, and belongs to the fields of traditional Chinese medicine and natural medicine. Background technique [0002] Pongamia pinnata (L.) Merr is a semi-mangrove plant of the genus Pongamia pinnata in the Fabaceae family. Literature data show that its whole plant can be used as medicine. "To treat scabies, burning ashes can also rub ringworm" ("Herbal Medicinal Preparations"); "The oil squeezed from the seeds can cure scabies, abscesses and rheumatism", "The whole plant of Shuiliu bean can be used as an emetic. "("Flora of Guangzhou"). The chemical composition of pumella is relatively complex. At present, domestic and foreign scholars mainly isolate flavonoids, triterpenes, alkaloids, amino acids and β-sitosterol and other compounds from it. According ...
