Embodiment Construction

[0075] The present invention is further described below in conjunction with the examples.

[0076] The high-sensitivity and high-specificity gene diagnosis and detection core reagent proposed by the present invention, which can be transported at room temperature, is a creative modification of the existing high-sensitivity and high-specificity gene diagnosis and detection core reagent. On the basis of the original reagent composition, by adding an appropriate amount of polysaccharide and disaccharide substances, the inventor not only explored the problem of maintaining the high sensitivity and high specificity of the original reagent, but also enabling the reagent to be transported at room temperature.

[0077] This highly sensitive and specific genetic diagnostic side core reagent that can be transported at room temperature includes reaction buffer, deoxyribonucleic acid triphosphate (DNTP), heat-resistant DNA polymerase, glycerol, bovine serum albumin (BSA), dithiothreonium S...

Abstract

The invention relates to a gene diagnosis and detection core reagent transportable at normal temperature and of high sensitivity and high specificity. The reagent comprises reaction buffer, deoxyribonucleoside triphosphate (DNTP), heat resistant DNA polymerase, glycerin, bovine serum albumin (BSA), dithiothreitol (DTT) and tween-20. The reagent is characterized in that: proper amounts of polysaccharide and disaccharide are mixed into the core reagent; the reaction buffer of the reagent is composed of 5mM-50mM of Tris-HCL, 20mM-500mM of KCL, 1.0mM-5mM of MgC12 and 20mM-200mM of (NH4)2SO4; meanwhile 50uM-300uM of DNTP, 0.01U/ul-0.2U/ul of heat resistant DNA polymerase, 1mM-15mM of polysaccharide, 2M-100M of disaccharide, 3%-30% of glycerin, 10-300ug/ml of BSA, 0.5-10mM of DTT and 0.005-0.05v/v of tween-20 are mixed.

Description

technical field [0001] The invention relates to a gene diagnosis detection reagent, in particular to a highly sensitive and high specificity gene diagnosis detection core reagent which can be transported at room temperature, and belongs to the gene diagnosis detection technology. Background technique [0002] PCR technology, that is, polymerase chain reaction (polymerase chain reaction, PCR) was established by Kary Mullis of PE Cetus Company in the United States in 1983 (won the Nobel Prize in Chemistry in 1993). This technology can amplify a specific DNA fragment millions of times after a few hours of reaction in a test tube. This technology of rapidly obtaining a large number of single nucleic acid fragments is of pivotal significance in molecular biology research and has greatly promoted Research progress in life sciences. It is not only the most commonly used technology for DNA analysis, but also has important application value in DNA recombination and expression, gene ...
