Embodiment Construction

[0026] The present invention will be described in more detail below through specific embodiments and with reference to the accompanying drawings. It should be understood that the examples described below are only for illustration rather than limitation of the present invention.

[0027] 1. Preparation of cDNA from Eucalyptus

[0028] Using the existing technical method adopted by Guo at al. 2008, fresh eucalyptus (Eucalyptus cv DH32-29) leaves were selected, frozen in liquid nitrogen, and then stored in a -80°C refrigerator for later use.

[0029] Use the Cetyltrimethyl Ammonium Bromide (CTAB) method to extract tissue total RNA. Specific implementation: 1) Preheat 15 ml CTAB extraction solution in 65°C water; 2) Grind in liquid nitrogen for 2-3 1 gram of eucalyptus frozen tissue; 3) Transfer the sample to a centrifuge tube with CTAB extract, vortex vigorously for 30 seconds immediately, and then bathe in water at 65°C for 4-5 minutes; 4) Add an equal volume of chloroform / iso...

Abstract

The invention discloses a WPGS 1 gene and a WPGS 2 gene derived from eucalyptus and encoding protein and application thereof and belongs to the field of biotechnology. Ribonucleic acid (RNA) in the eucalyptus is extracted and is subjected to reverse transcription to be complementary deoxyribonucleic acid (cDNA), deep sequencing is performed to obtain an overall length cDNA sequence of the eucalyptus, the similarity between a deoxyribonucleic acid (DNA) sequence and the eucalyptus cDNA sequence library sequence obtained through sequencing is compared to obtain overall length cDNA sequences of the WPGS 1 gene and the WPGS 2 gene in the eucalyptus and lead the sequences into Arabidopsis, wherein the DNA sequence is presumed according to AtGA-20 and AtEXO protein amino acid sequences of arabidopsis published by center of biotechnology information (NCBI). The biomass of transgenic plants is remarkably improved through over-expression of the WPGS 1 gene and the WPGS 2 gene.

Description

technical field [0001] The present invention relates to plant gene and coded protein and application, particularly relate to two originating from eucalyptus [0002] WPGS1 gene and WPGS2 gene and their coded proteins and their application in improving plant biomass. Background technique [0003] Eucalyptus is the general name of Eucalyptus in the family Myrtaceae. It is an excellent tree species that integrates ornamental, timber, pulp raw materials, and medicinal uses. Due to the long growth cycle of forest trees and high genetic heterogeneity, many traits are quantitative traits controlled by polygenes. Conventional breeding methods are difficult to meet the requirements of directional breeding of new eucalyptus varieties. Therefore, people place their research focus and success on genetic engineering technology. Solve the difficult problems of conventional eucalyptus breeding, and accelerate the process of breeding new varieties of high-quality and high-yielding eucalypt...
