Problems solved by technology

However, prolonged hydrolysis is generally not a desirable effect in depilling as it may result in damage to the fabric or severe strength loss
Furthermore, hydrolysis may not correlate with biopolishing effect as reported by Ramos et al. (Biocatalysis and Biotransformation, 2007, 25(1):35-42)

Embodiment 11

[0147] Example 1.1: Host Trichoderma reesei strains for overexpression of Cel45A cellulase

[0148] The parent Trichoderma reesei strain used to overexpress the TrCel45A cellulase was P998A (described in co-pending and commonly owned US Patent Application No. 61 / 312,864). Strain P998A was generated by transforming the BTR213 uridine auxotroph strain with a transformation vector (Pc / x-cel5A-G363A-pyr4-TV) designed to express high levels of Cel5A- G363A (SEQ ID NO: 1) enzyme and restores prototrophic growth in the absence of uridine (see US Patent Application No. 61 / 312,864, supra). Strain BTR213 is a derivative of strain RutC30 (ATCC #56765) which was isolated as a high cellulose expressing derivative of progenitor strain QM6A (Montencourt et al., Proc. 3rd Annual Symp. On Fuels from Biomass, Golden Colorado, 1979, pp. 85-89). Further cellulase hyper-producing strains were generated from RutC30 by random mutation and / or selection. Bacterial strain M2C38 (ATCC74252) is based ...

Embodiment 12

[0149] Example 1.2: Transformation vector of Trichoderma reesei for overexpression of TrCel45A

[0150] Three TrCel45A expression vectors were used for T. reesei transformation. All vectors were designed to target the native Trcel7a locus. To facilitate targeting, sequences amplified from BTR213 genomic DNA adjacent to the 5' and 3' ends of the native Trcel7a gene were inserted into the transformation vector such that they flanked the expression and selection cassettes ( Figure 4-6 ).

[0151] Transformation vector pP7a-TrCel45A-hph-TV ( Figure 4 ) contains the E. coli hygromycin phosphotransferase gene (hph) as a selectable marker. The transcription of the Hph gene is driven by the Trichoderma reesei phosphoglycerate kinase promoter (Ppgk) and is mediated by the Neurospora crassa orotidine-5'-monophosphate-decarboxylase (Tpyr4) gene (pyr4) terminated by the terminator. The expression cassette contains the Trichoderma reesei cel7a promoter (Pcel7a) as part of the 5' fla...

Abstract

The present invention relates to a depilling composition comprising an enzyme mixture that comprises a Family 45 cellulase enzyme component and one or more additional cellulase enzyme components selected from a Family 5 cellulase, a Family 6 cellulase or a combination thereof. The enzyme mixture may be characterized in that the Family 45 and the Family 5 cellulase enzyme components or the Family 45 and the Family 6 cellulase enzyme components are present at a weight ratio that exhibits synergy in an assay that measures specific depilling activity. The enzyme mixture may be secreted by a genetically modified microbe overexpressing theforegoing cellulase enzyme components. Also provided is a process for depilling that comprises a step of contacting cellulose-containing goods with the depilling composition.

Description

field of invention [0001] The present invention relates to cellulase mixtures and uses thereof, in particular cellulase mixtures for the treatment of cellulose-containing articles. Background of the invention [0002] Cellulases are widely used to improve the appearance and softness of cellulose-containing fabrics. One widespread use of cellulases is to remove cotton fuzz and loosen surface fibers in or on fabrics. This process is called "depilling", "biopolishing", and "biofinishing", and it smoothes the surface of the fabric, which in turn improves its softness and appearance. Cellulase treatment also helps prevent the subsequent formation of fiberballs that make clothes look worn. During depilling, it is desirable to minimize the loss of strength of the fabric due to the hydrolysis of cellulase enzymes. [0003] Another industrial application of cellulases is in the treatment of denim fabrics to give them a "stone-washed" appearance. This type of process is called "bi...
