Three segments of relatively conservative nucleotide sequences are obtained by analyzing the sequences of 10 known avian IFN-alpha genes, and based on the three segments of sequences, three degenerate primers for cloning avian alpha-interferon genes are designed and synthesized, wherein the nucleotide sequence of one upstream degenerate primer AIF-TY1a is 5'-CASCRSYACRBCCASCASCTCSAGC-3', while the nucleotide sequence of the other upstream degenerate primer POAU is 5'-CYRSYSCTCNYGCTCCTYCT-3', and the nucleotide sequence of the common downstream degenerate primer AIF-TY2b of the two upstream degenerate primers is 5'-AGGMGGACGRGKTCCCASGCGCAG-3', wherein R=A / G, Y=C / T, M=A / C, K=G / T, S=C / G, and B=C / G / T.
