Embodiment

[0021] 1. Blood sample collection

[0022] Collect 5-10mL of blood from the jugular vein of the sheep after Baoding, put it in a 15mL finger tube (with 1-2.5mL of lysate inside), and shake it well until the blood cells are completely ruptured and the solution becomes viscous.

[0023] 2. Primer information

[0024] Using Primer5.0 software, primers were designed according to the relevant OPRK1 gene sequences NC_007312 and NW_00149302 included in GenBank (Table 1). All primers were synthesized by Shanghai Shenggong Technology Co., Ltd. The primer dilution process is carried out in 2 steps: first, dilute to 10 times the concentration of normal amplification, and store at -20°C for a long time; then dilute 10 times when used, as the normal primer concentration.

Abstract

The invention belongs to the technical field of biological detection and relates to a method and a reagent for detecting the stereotyped behavior of a sheep by virtue of the single nucleotide polymorphism of an OPRK1 (Kappa Opoioid Receptor) gene. With the PCR-SSCP technique as a means, the single nucleotide polymorphism T118A or G90A of the OPRK1 gene in the sheep is discovered, and the polymorphism of the gene is associated with stereotyped behavior traits of the sheep, and by virtue of the detection, whether the sheep suffers from the stereotyped behavior is found as soon as possible and as quickly as possible without the need of long-time observation on animals and need of necessary adjuvant therapy, and thus the method is simple and feasible, the production loss is reduced and the production capacities of animals are improved.

Description

technical field [0001] The invention belongs to the technical field of biological detection, and relates to a method and a reagent for detecting behavior of sheep behavior by using OPRK1 gene SNP. Background technique [0002] Some studies have determined that pigs stand still for a long time as a kind of animal "rule behavior". Under captive conditions, animals lying down or standing and watching for a long time can be considered as a kind of "regular addiction" or "stiff" behavior of animals. Some scholars believe that the appearance of behavioral stigma is caused by over-selection of production traits, and some scholars have found that stigma is caused by the monotonous and narrow living environment of animals in captivity. Grandin (1998) did not find tongue rolling and other abnormal behaviors in cattle in the 1970s, but in 1996 it was found that Holstein cattle breeds showed tongue rolling, which may be the result of long-term directional selection. High-producing sows...
