Problems solved by technology

At present, many patients with bacterium-negative pulmonary tuberculosis have no specific clinical manifestations and lack typical imaging features. Therefore, it is easy to miss and misdiagnose tuberculosis with existing tuberculosis diagnostic methods, which is not good for prognosis, and it is easy to cause tuberculosis to be difficult to control or spread.

[0009] Objects and Methods

[0010] 1. Objects: From January 2012 to December 2012, 60 cases of bacteria-negative pulmonary tuberculosis were admitted to Wenzhou Medical University's Ding Li Clinical College, and 20 cases of outpatient health screening were conducted during the same period. All enrolled subjects had no medical history of immunodeficiency virus infection, acute virus infection, pregnancy and application of immunosuppressants.

[0011] 1. Experimental group: 60 patients with bacterial negative pulmonary tuberculosis, 43 males and 17 females. Aged 20-52 years old, with an average of 35.4±15.3 years old. Among them, there were 21 cases with bacteriological or pathological evidence and tuberculosis infection in other parts, including 2 cases of tuberculous meningitis, 4 cases of tuberculous pleurisy, 1 case of renal tuberculosis, 2 cases of tuberculous peritonitis, 3 cases of joint tuberculosis and lymphatic tuberculosis. 9 cases of tuberculosis; 39 cases of pat...

Abstract

The invention discloses an enzyme-linked immunospot test method for T lymphocytes. The method comprises the following steps: by adopting a T-SPOT.TB kit produced in British Oxford Immunotec Company, taking 5ml of sodium citrate anticoagulation venous blood, separating mononuclear cells of peripheral blood and placing the mononuclear cells on a gamma-interferon antibody coated porous plate, adding an early secretory antigenic target (ESAT-6) of tuberculosis specific antigen and a culture filtrate protein (CFP-10) to serve as stimulus antigens, culturing in an incubator for 24 hours, washing off T lymphocytes with antigen sensitization effects, adding a biotin labeled second antibody, performing enzyme linked colorimetry, forming spots at secreting positions of cell factors, and detecting the number of the T lymphocytes with the tuberculosis specific antigen sensitization effects by recording the spot number. The method disclosed by the invention contributes to early screening of culture negative pulmonary tuberculosis.

Description

technical field [0001] The invention belongs to the field of biological detection, in particular to a T cell spot test method. Background technique [0002] In recent years, the incidence of tuberculosis has been on the rise. Bacterial-negative pulmonary tuberculosis accounts for 40% to 60% of all pulmonary tuberculosis. If patients with bacterial-negative pulmonary tuberculosis are not treated, nearly half of the patients will develop smear-positive and become a new source of infection. Contents of the invention [0003] The purpose of the present invention is to overcome the shortcomings and deficiencies of the pr...
