Problems solved by technology

However, most of the studies are limited to a certain type, especially the detection of teicoplanin, and are aimed at body fluid (blood) samples, and only vancomycin and norvancomycin are used for animal-derived food pork. and detection of teicoplanin
The simultaneous detection of vancomycin, teicoplanin, telavancin, dalbavancin, oritavancin in foods of animal origin has not been reported

Embodiment 1

[0028] A method for the simultaneous detection of five glycopeptide antibiotics in food of animal origin, among which the five glycopeptide antibiotics telavancin, teicoplanin, oritavancin, dalbavacin, vancomycin Schematic diagram of the structure of vancomycin figure 1 As shown, it mainly includes the following steps:

[0029] Weigh 5g of homogeneous sample, put it in a 50mL polypropylene centrifuge tube, add 20mL 0.1mol / L EDTA-Mcllvaine buffer solution, vortex mix at 1000r / min for 1min, extract on a shaker for 15min, then add 10mL methanol to precipitate protein for 10min , and then use a refrigerated centrifuge to centrifuge at 10,000 r / min for 5 minutes (the temperature is lower than 5° C.), take the supernatant, filter it with fast filter paper, and wait for purification.

[0030] Connect the HLB solid-phase extraction cartridge (60mg, 3mL) and the MCX solid-phase extraction cartridge (60mg, 3mL) in series with an adapter, activate with 3mL of methanol and equilibrate wi...

Embodiment 2-7

[0049] Separately adopt solid-phase extraction columns HLB, C18, MCX, WCX, WAX, MAX to replace the HLB solid-phase extraction column and strong cation (MCX) solid-phase extraction column series extraction in Example 1. The obtained comparative results are as follows Figure 4 shown.

Abstract

The invention discloses a method for simultaneously detecting various glycopeptides antibiotics in animal derived food. The method mainly comprises the following steps: carrying out solution extraction on a sample; carrying out series purification and enrichment by using HLB solid-phase extraction columns and strong cation (MCX) solid-phase extraction columns; separating the sample by using reversed phase columns, and then detecting the sample by using an ultra-efficient liquid chromatography tandem mass spectrometry in a multi-reaction monitoring mode; and carrying out qualitative and quantitative analysis. Five glycopeptides antibiotics including telavancin, teicoplanin, oritavancin, dalbavancin and vancomycin in the sample animal derived food such as milk, eggs and chicken can be detected simultaneously. The detecting method is sensitive, accurate and rapid; and the five glycopeptides antibiotics can be qualitatively and quantitatively analyzed by six minutes displayed on an instrument. Moreover, the detecting method also has certain reference value for detection of other glycopeptides antibiotics.

Description

technical field [0001] The invention belongs to the field of antibiotic detection, in particular to a simultaneous detection method for multiple glycopeptide antibiotics in animal-derived food. Background technique [0002] my country is a big country in the production and use of antibiotics. The irrational use of antibiotics in clinical practice, antibiotic pollution in the water environment and antibiotic residues in food have led to the emergence of drug resistance in many pathogenic bacteria. On the one hand, the increase of drug-resistant bacteria has caused the further abuse of commonly used antibiotics, and on the other hand, it has also put forward higher requirements for antibiotic products clinically. Due to the marketed use of penicillin and cephalosporin antibiotics, glycopeptide antibiotics are reserved drugs and are usually used clinically as a last resort after failure of β-lactam antibiotics or other antibacterial drugs; considered to be The last line of def...
