Problems solved by technology

[0004] Apis cerana cerana (abbreviated as Apis cerana) is a subspecies of Apis mellifera. It is a unique bee species in China. It has the advantages of using sporadic nectar source plants, strong collection ability, high utilization rate, long honey collection period and adaptability. Strong anti-mite and disease resistance, less feed consumption and other incomparable advantages of Italian bees, it is very suitable for fixed-point breeding in mountainous areas of China, but its honey production is low, and it does not produce propolis and royal jelly
[0005] Zhongfeng honey is also called "earth honey". Its nutritional and health benefits are deeply trusted by consumers. However, due to the low output, its market price is several times that of ordinary Italian bee honey
Some unscrupulous vendors used Italian bee honey to pretend to be or mixed with Chinese bee honey to earn high profits, which caused great losses to Chinese bee farmers and related bee product companies, and at the same time triggered a crisis of trust among consumers. negative impact on the brand image of the bee product market, which is not conducive to the healthy development of the bee product market

Embodiment 1

[0031] A method for discriminating Chinese bee honey and Italian bee honey is realized through the following steps:

[0032] (1) Extraction of alkane components in honey: Weigh 20g of honey, put it in a 50mL centrifuge tube, add 20mL of pure water, vortex and mix well, centrifuge at 15000g for 10min at room temperature, discard the supernatant, add petroleum ether 60-90 to precipitate ℃ 10mL, vortex shake for 10-15min, centrifuge at 8000g for 10min at room temperature, pipette the supernatant into a 15mL centrifuge tube to obtain honey extract H1.

[0033] (2) Concentration of the honey extract: Concentrate the honey extract H1 with a nitrogen blowing instrument, control the heating temperature of the nitrogen blowing instrument at 30-40°C, concentrate the extract to 1-2mL, stop the nitrogen blowing, and obtain the extract H2 .

[0034] (3) Preparation of n-triaconane standard working solution (1mg / mL): Weigh 50mg of n-triaconane standard product into a 50mL volumetric flask,...

Embodiment 2

[0050] In order to ensure the experimental effect, the raw honey in this example comes from several cooperative bee farms in Jinhua City, Zhejiang Province and the experimental bee farm of Zhejiang University. The sample information is shown in Table 1 below.

[0051] Table 1 Honey sample information table

[0052]

[0053] Carry out experiment according to the method step of embodiment one, gas chromatography result sees figure 1 , it can be clearly seen in the figure that there is a clear difference in the peak diagrams of the two honeys. Italian bee honey has a characteristic peak at 21.3 minutes, and Zhongfeng honey has a characteristic peak at 29.2 minutes. The subsequent mass spectrometry results show that Italian bee honey has a characteristic peak at 21.3 minutes. The characteristic peak components are C31 alkane components, and the characteristic peak components of Zhongfeng honey at 29.2min are C37 alkane components. Honey from two different bee species can be id...

Embodiment 3

[0055] The honey samples in this example come from several cooperative bee farms in Jinhua City, Zhejiang Province and the experimental bee farm of Zhejiang University. Two honey samples from Chinese bees and two honey samples from Italian bees are blended. The blending ratio of the two kinds of honey is 50%, the number of blended honey samples is 2. Carry out experiment according to the method step of embodiment 1, blend honey gas chromatography result see figure 2 . from figure 2 It can be clearly seen that there are two characteristic peaks of C31 and C37 alkanes in the blended honey, which proves that the honey sample is blended honey.

Abstract

The invention provides a method for identifying the honey of Apis cerana and the honey of Apis mellifera. The method achieves the purpose of identifying the two kinds of honey based on the differenceof alkane components in the two kinds of honey. Honey samples are extracted with petroleum ether and then detected via gas chromatography-mass spectrometry; and detection results show that the alkanecomponents of the honey of Apis cerana and the honey of Apis mellifera are different, the honey of Apis cerana contains C37 alkanes, and the honey of Apis mellifera contains C31 alkanes. It is confirmed that the component difference is only related to bee species after detection and analysis of a large number of samples, and the influence of plant sources on identification is excluded. The differential alkane components are stable in honey and are not affected by concentration processing and shelf life. The method of the invention is reasonable in design, convenient, quick and accurate in identification, and is applicable to both the identification of the sources of raw honey and commercial bee species and the identification of the honey of Apis cerana blended with a certain proportion ofthe honey of Apis mellifera.

Description

technical field [0001] The invention belongs to the field of bee products, and mainly relates to a method for identifying Chinese bee honey and Italian bee honey, which is to identify Chinese bee honey and Italian bee honey by means of instrument analysis. technical background [0002] Honey is a natural sweet substance that bees collect nectar, secretions or honeydew from plants, combine with their own secretions, transform, dehydrate, store and mature in the honeycomb. The secretions of bees in honey are mainly some proteins, including some digestive enzymes and royal jelly protein. [0003] Italian honey bee (Apis mellifera ligustica, referred to as Italian honey bee) is a subspecies of Western honey bee, native to Italy, with strong ability to produce honey and royal jelly. Ideal variety, also used for propolis production. It is a bee species widely cultivated in the world, and it is also the main species of bee culture in China. [0004] Apis cerana cerana (abbreviat...
