Problems solved by technology

Although strict protocol-based treatment is a key factor in reducing hospital stays (Hall ES et al (2014) Pediatrics Vol 134:e pp 527-534), national standards governing treatment withdrawal strategies are lacking
The general goal of existing strategies is to alleviate the withdrawal response only after the neonate has already developed dependence, and there is a lack of prenatal strategies to prevent the development of neonatal dependence in the first place

[0081] The C57BL / 6NTac strain of mice was produced from a breeding colony in Ohio. Animals were housed in well-ventilated microisolation cages with 24-hour access to food and water. Put them on a 12h:12h circadian cycle. All procedures were approved by The Ohio State University Institutional Animal Care and Use Committee and in accordance with guidelines established by the National Institutes of Health and published in the "Guide for the Care and Use of Laboratory Animals". Abstract

Disclosed is a method for reducing or preventing fetal opioid dependence in a drug dependent or opioid tolerant pregnant subject. The method involves administering to the pregnant subject a composition comprising an opioid antagonist in an amount effective to reduce or prevent fetal opioid dependence, wherein the opioid antagonist a) is orally available or delivered systemically and reaches the circulation of the pregnant subject, b) is substantially excluded from the subject's brain by the blood brain barrier, and c) penetrates the placenta and enters the fetal brain. The method also includesadministering the opioid antagonist to the neonate in increasing amounts to facilitate weaning the neonate from continued opioid maintenance.

Description

[0001] Cross References to Related Applications [0002] This application claims the benefit of US Provisional Patent Application Serial No. 62 / 276,691 filed January 8, 2016, the disclosure of which is expressly incorporated herein by reference. technical field [0003] The present invention relates to compounds, compositions and methods for the treatment and prevention of opioid neonatal withdrawal syndrome. Background technique [0004] Illicit drug use by pregnant women is a growing national problem estimated to be present in about 16% of teenage pregnant women and about 7% of women aged 18 to 25 (Patrick SW et al. (2012) JAMA Vol. 307: 1934 to 1940). The estimated prevalence of infants and young children born with opioid dependence manifested as neonatal withdrawal syndrome (NAS) has reached 3.39 per 1,000 births as of 2009 (Patrick SW et al. (2012 2007) JAMA Vol. 307: pp. 1934-1940). A significant proportion of such newborns are mothers who are receiving managed meth...
