Problems solved by technology

Paclitaxel and cisplatin are chemotherapy drugs commonly used in ESCC, however the response rate of both drugs is as low as 33-40%, and in about 50% of patients, the tumor recurs rapidly even after several rounds of chemotherapy and the prognosis is poor

Embodiment Construction

[0013] The present invention will be described in further detail below in conjunction with examples and specific implementation methods.

[0014] 1) SOX15 is overexpressed in ESCC

[0015] According to the existing research results, SOX15 is significantly overexpressed in the early stages of human primordial germ cell development, and its expression is reduced in later stages, suggesting that SOX15 may play an important role in early embryonic / germline development. Like other transcription factors such as OCT4 and cKIT, their reactivation in adult tissues may be involved in tumorigenesis and progression.

[0016] In order to determine the role of SOX15 map in tumors, according to the expression of SOX15 gene and DNA mutation status in TCGA database, the expression of SOX15 in esophageal adenocarcinoma, breast cancer, colon / rectal adenocarcinoma, ovarian cancer, prostate cancer, skin melanoma, It was reduced in a subset of endometrial carcinomas and uterine carcinosarcomas; co...

Abstract

The invention relates to application of an SOX15 gene serving as a biomarker for guiding drug therapy of ESCC (esophageal squamous cell carcinoma). Drug therapy involves a drug namely paclitaxel. TheSOX15 gene serving as the biomarker for guiding drug therapy of ESCC suppresses SOX15 gene expression in an ESCC cell line to suppress cell proliferation, so that cancer cells are sensitive to ESCC drug therapy. The application has advantages that sensitivity of the cancer cells to ESCC paclitaxel therapy is realized through cell proliferation suppression resulted from SOX15 expression suppression; due to SOX15 gene silencing, gene expression in WNT and NOTCH signal transduction is reduced; the SOX15 gene can be used as a potential novel biomarker for guiding ESCC therapy with paclitaxel; effects of ESCC therapy with paclitaxel can be predicted; remarkable assistance is provided for ESCC therapy.

Description

technical field [0001] The present invention relates to the application of inhibiting the expression of SOX15 gene, in particular to an application of inhibiting the effect of the expression of SOX15 gene on the sensitivity of ESCC cells to paclitaxel treatment. Background technique [0002] ESCC (esophageal squamous cell carcinoma) is the major tissue type of esophageal malignancy in China. The landscape of genomic alterations in ESCC has been well characterized by multiple studies. Frequent amplification of DNA sequences containing CCND1 and TP63 has been found, And most of the gene mutations in TP53, NFE2L2, MLL2, ZNF750, NOTCH1 and TGFBR2 have been previously identified in ESCC. Paclitaxel and cisplatin are chemotherapy drugs commonly used in ESCC, however the response rate of both drugs is as low as 33-40%, and in about 50% of patients, the tumor recurs rapidly even after several rounds of chemotherapy and the prognosis is poor . Therefore, research on the chemical re...
