Embodiment 1

[0042] [Example 1] Preparation of bacteria-carrying composite microspheres

[0043] 1. Preparation of degradative strain PB-1 bacterial suspension

[0044] 1.1 Bacteria picking: Use a toothpick to pick a single colony on an LB plate stored at 4°C, place it in a 2mLEP tube containing 1mL of sterilized LB, and incubate at 180rpm at 30°C for 14h until the culture medium becomes turbid to activate and degrade the strain .

[0045] 1.2 Transformation and cultivation: expand the cultivation of degraded strains, add the activated degraded strains into the Erlenmeyer flask filled with LB according to 1% (V / V), cultivate under the same conditions for 16 hours, and the culture medium presents an orange-red turbid liquid.

[0046] 1.3 Bacteria collection: Pour the cultured bacteria in step (2) into a sterilized and dry 50mL centrifuge tube, centrifuge at 6000rpm for 10min, remove the supernatant, and wash the bacteria three times with an appropriate amount of MSM medium. Finally, the b...

Abstract

The invention discloses a bacteria-loaded composite microsphere for synchronously removing phenol and aniline, its preparation method and application. Film formation, the degrading strain belongs to the genus Rhodococcus, named Rhodococcus sp.PB‑1, and is preserved in the China Center for Type Culture Collection with the preservation number CCTCC NO.M2019472. In the present invention, the optimum bacteria content, sodium alginate, bamboo charcoal and chitosan concentrations are determined through orthogonal experiments. The bacteria-loaded composite microsphere provided by the invention has remarkable effect on removing phenol and aniline, can be recycled, has simple preparation method and low production cost, and has very broad application prospect in treating industrial wastewater polluted by related organic matter.

Description

technical field [0001] The invention belongs to the field of biological material preparation, in particular to a bacteria-carrying composite microsphere for synchronous removal of phenol and aniline, its preparation method and application. Background technique [0002] Among aromatic compounds, phenol and aniline have good reactivity and high product efficiency, and are commonly used raw materials or intermediates for the synthesis of fine chemicals, agrochemicals, pharmaceuticals, and dyes, and thus become the main harmful pollutants in the wastewater discharged by related industries composition of matter. Among the various organic pollutants, phenol and aniline are among the toxic and persistent class, mutagenic and potentially lethal. Phenols are readily absorbed through skin contact and may act as potent endocrine disruptors; the primary toxicity of anilines is binding to hemoglobin in the blood, reducing oxygen transport. Both phenol and aniline are listed as priority...
