Embodiment Construction

[0025] The present invention will be further described below in conjunction with embodiment. The examples are intended to illustrate the present invention, but not to limit the present invention in any form.

[0026] 1. Provide tissue samples of Tridacna safranata in accordance with the sample submission requirements of Nuohezhiyuan Bioinformation Technology Co., Ltd. without reference. Select 5 individuals with an average shell height of 7±1 cm from safflower clam, take 0.1 g each of the mantle, obturator muscle, gonad, and lymphocytes or tissues, add Trizol (Invitrogen, USA), and grind with a tissue grinder , put it into liquid nitrogen and freeze immediately, and deliver it to the company by dry ice express the next day, and carry out the next step of experimental processing according to each tissue sample, including RNA extraction, quality inspection, transcription, library construction, and Illumina HiSeq sequencing. The simple process is as follows figure 1 .

Abstract

The invention discloses a giant clam immune-related SAA gene and its application in preparing a giant clam pathological detection reagent. The present invention clones the SAA gene from Tridacna safranata for the first time, and designs general primers TcSAA‑F / TcSAA‑R for the gene in several common Tridacna. Quantitative PCR experiments showed that the SAA gene of Tridacna safranatum was significantly up-regulated after Vibrio infection, which was very similar to that of mammals. In other pathological conditions, the expression of this gene was also significantly up-regulated. Therefore, the SAA gene can also be used as a gold standard for detecting the health status of giant clams. The SAA gene and primers TcSAA-F / TcSAA-R of the present invention can be used to monitor the health status of giant clam, provide a positive and accurate warning signal for the prevention of giant clam disease, and can effectively prevent the expansion of disasters. The application of this technology will further enhance the ability of giant clam breeding germplasm innovation and support the efficient and sustainable development of the industry.

Description

Technical field: [0001] The invention belongs to the technical field of molecular biology, and in particular relates to a SAA gene related to immunity of giant clams and its application in preparing reagents for detecting the pathology of giant clams. Background technique: [0002] Tridacna belongs to the phylum Molluscs, Phyllobranchia, Pleurobranches, and Tridacnaidae. There are 2 genera. There are 9 species in the genus Tridacna, and only 1 species in the genus Tridacna. Tridacna is the largest shellfish today, and some species can reach about 1 meter in size and weigh more than 300 kilograms. In addition to being the largest in size, Tridacna can also be self-supporting, because there are symbiotic zooxanthellae in its mantle , can carry out photosynthesis under light conditions, provide nutrients for giant clams, enable them to survive in the open sea where nutrients are extremely deficient, and provide the cornerstone for the construction of coral reefs. Tridacna are ...
