Embodiment 1

[0029] 1. Primer design and synthesis: According to the mitochondrial sequences of Melonid flies and South Asian fruit flies in the NCBI database, the online program Primer-BLAST (https: / / blast.ncbi.nlm.nih.gov / Blast.cgi) was used to design the upstream and downstream of HRM Primers F and R, where the sequences of primers F and R are:

[0030] Primer F: 5'-CCTGGTCCTTTCGTACTAAGGTATT-3' (SEQ ID No. 1);

[0031] Primer R: 5'-TGATCTGAGTTCAGACCGGC-3' (SEQ ID No. 2).

[0032] 2. Extraction of DNA from fruit flies and fruit flies from South Asia: Using commercially available genomic DNA extraction kits (Tiangen Biochemical Technology (Beijing) Co., Ltd., product model: DP304), single-head individuals of fruit flies and fruit flies from South Asia were extracted respectively. Genomic DNA, micro-UV spectrophotometer NanoDrop-1000C Thermo, USA) detected the quality and concentration of extracted DNA.

Abstract

The invention discloses an HRM primer, a kit and a method for rapidly identifying bactrocera cucurbitae and South Asia fruit flies. The method comprises the steps of firstly, designing and synthesizing a HRM specific primer for distinguishing two kinds of fruit flies based on mitochondrial genomes of bactrocera cucurbitae and South Asia fruit flies; respectively extracting genome DNAs of bactrocera cucurbitae and South Asia fruit flies; carrying out high-resolution real-time fluorescent quantitative PCR analysis by taking the extracted DNAs as templates and EvGreen as a dye; and distinguishingtwo types of bactrocera cucurbitae according to peak difference of melting temperature curves. According to the invention, the problem of rapid identification of two kinds of bactrocera cucurbitae and South Asia fruit flies which are co-hosted and have similar larvae is solved; the HRM method for identifying the two kinds of bactrocera cucurbitae based on mitochondrial genes is established; bactrocera cucurbitae and South Asia fruit flies which are mixed on melon vegetables can be quickly, efficiently and simply distinguished; and the HRM primer, the kit and the method have the advantages ofhigh accuracy, high flux, good repeatability and low cost.

Description

technical field [0001] The invention belongs to the technical field of insect molecular biology, and in particular relates to HRM primers, a kit and a method for rapidly identifying the fruit fly and the fruit fly of South Asia. Background technique [0002] Bactrocera cucurbitae and Bactrocera tau belong to the genus Bactrocera of Tephritidae, Diptera, and are important pests in the production of melons, fruits and vegetables in Southwest my country and South China. and quarantine organisms. The adults lay eggs in the host fruit, and the hatched larvae feed on the host tissue in the fruit, causing decay and fruit drop, affecting the yield and quality of fruits and vegetables, and causing huge economic losses. The prevention and control of the above-mentioned two kinds of fruit fly damage is of great significance for improving the yield and quality of fruits and vegetables and ensuring agricultural economic income. [0003] Prediction of pest occurrence is an important part...
