Abstract

The invention discloses a mutant enzyme of type I pullulanase. An amino acid sequence of the mutant enzyme is shown as SEQ ID No. 1. The invention also discloses a nucleotide sequence of a gene for coding the mutant enzyme as shown in SEQ ID No. 2. The invention also discloses a preparation method of the mutant enzyme, which comprises the following steps: carrying out homologous modeling accordingto the amino acid sequence of type I pullulanase, selecting mutation sites based on sequence alignment analysis and protein structure analysis, and designing mutation primer pairs; respectively carrying out site-specific mutagenesis reaction by taking a carrier carrying the I-type pullulanase gene as a template to obtain mutant plasmids; and copying and expressing the corresponding mutant enzymein the mutant plasmid transformation engineering bacteria. The mutant enzyme provided by the invention has better heat resistance and specific enzyme activity.
