Abstract

The invention relates to the field of yeast biotechnology and synthetic biology, in particular to a phaffia rhodozyma RNA polymerase III promoter and application thereof. The promoter is a phaffia rhodozyma RNA polymerase III promoter; and the sequence of the promoter is SEQ ID NO:1, SEQ ID NO:2, SEQ ID NO:3 or SEQ ID NO:4, or a DNA nucleotide sequence which has 70-percent homology with SEQ ID NO:1, SEQ ID NO:2, SEQ ID NO:3 or SEQ ID NO:4 and has promoter activity. The promoter disclosed by the invention is applied to a phaffia rhodozyma CIRSPR-Cas9 system for guiding the gRNA expression of the system, and the gene editing efficiency is improved by 4 times.
