[0285] The heavy chain variable domain (VH) and light chain variable domain (VH) cDNAs of MCA44-3A6 were cloned using oligonucleotide primers available for cDNA synthesis and PCR methods. Clones were based on sequences published in the Immunoglobulin Database. Briefly, with commercially available reagents, from about 5x10 8 RNA prepared by hybridoma cells was used to prepare cDNA. The amplified cDNAs of VH and VL were digested with restriction enzymes whose sites were installed during PCR amplification. The genes were then subcloned into phage display vectors to construct scFvs for library selection purposes.
