Embodiment 1

[0068] All patients were from the Department of Dermatology, Huashan Hospital Affiliated to Fudan University, Shanghai, China. The Ethics Committee of Huashan Hospital approved the protocol, and all patients and healthy controls provided written informed consent.

[0069] Eight PsA patients and 15 PsO patients were recruited for exosomal miRNA sequencing. The diagnoses of PsO and PsA were based on typical clinical and / or histopathological criteria and classification criteria for psoriatic arthritis (CASPAR), respectively. All PsA patients met CASPAR diagnostic criteria based on monthly follow-up assessments for at least one year. All patients were interviewed and data were recorded on disease duration, age of onset, smoking status, alcohol consumption, and medical history (including hypertension and diabetes). Two board-certified dermatologists graded skin psoriasis using the Psoriasis Area and Severity Index (PASI) and Body Surface Area (BSA) scores.

Embodiment 2

[0073] The miRNA in peripheral blood exosomes was collected using the method of Example 1, using the reverse transcription neck loop primer (SEQ ID NO: 2) and amplification primers (SEQ ID NO: 3 and 4) for miR-191-3p miRNA detection by RT-PCR. The results were similar to Example 1, the amount of miR-191-3p in patients with psoriatic arthritis was only 1 / 50 of that in patients with psoriasis vulgaris (p<0.001).

Abstract

The invention provides a miRNA marker related to psoriatic arthritis, a reagent for detecting the miRNA marker and application of the miRNA marker.

Description

technical field [0001] The invention belongs to the field of molecular auxiliary diagnosis, and specifically relates to the application in the diagnosis of psoriatic arthritis. Background technique [0002] Psoriatic arthritis (PsA) affects up to 30% of patients with psoriasis, resulting in decreased quality of life for patients. PsA involves multiple sites, including peripheral and axial arthritis, enthesitis, dactylitis, and nail disease. Early intervention in psoriatic arthritis is important to prevent loss of function and permanent disability. However, skin symptoms often preceded arthritis, delaying the diagnosis of PsA by an average of 5 years. Therefore, it is important for dermatologists to detect biomarkers that can be used to detect early warning patients of psoriatic arthritis and distinguish them from psoriasis vulgaris (PsO). [0003] In recent years, there has been growing interest in the importance of circulating microRNAs (miRNAs) carried by extracellular ...
