Problems solved by technology

The method of smut resistance identification affects the effectiveness of disease-resistant breeding. So far, domestic sugarcane varieties have been identified for smut resistance by manual inoculation and field planting identification methods. For the disease rate, the fixed 1-9 classification method is used to classify the resistance. After harvesting, the perennial root is continued to be observed, and finally evaluated and graded. The current method is greatly affected by the interaction of the environment, is not easy to control, and the test operation is complicated. The grading standard is not easy to grasp, and the results of the identification are quite different from the field resistance, which cannot accurately reflect the disease resistance of the variety

Embodiment Construction

[0014] Method steps of the present invention are:

[0015] 1. Screening standard control varieties

[0016] 1. Select a set of standard control varieties through at least 3 experiments.

[0017] 2. Include at least two highly resistant, two moderately resistant and two susceptible standard control varieties in the test.

[0018] 2. Artificial impregnation inoculation identification test

[0019] 1. Collect the spores of sugarcane smut.

[0020] a. Collect the diseased part, cut it off 10-20cm below the diseased part, remove the green leaves, put it in a bag, and only collect the part with many spores.

[0021] b. Put the collected diseased parts in a relatively low-temperature, dry, but non-freezing place, and cover them with plastic or wax paper.

[0022] c. Let them fully dry for 1-2 weeks until completely dry, which is necessary to maintain the survival of the spores. Spores must be stored in an environment with very low humidity.

[0023] d. Collect the spores by sie...

Abstract

The invention provides a method for identifying smut resistance in sugarcane. The method comprises the following steps that: screening a set of standard contrast varieties through at least three tests, which comprises at least two high resistance varieties, two medium resistance varieties and two infected standard contrast varieties; carrying out an artificial impregnation grafting evaluation test on the standard contrast varieties and materials participating in test evaluation simultaneously; recording the number of seedling emergence after the sugarcane is emerged; calculating the number of total clumps and the number of infected clumps on each line in harvesting; remaining perennial roots, and recording the number of seedling emergence and calculating the number of total clumps and the number of infected clumps on each line, and evaluating the clump infection rate according to the number of the total clumps and the number of infected clumps to calculate the infection rate; and calculating and evaluating the clone for classification through a regression equation of infection standard percentage (X) and standard classification thereof (Y). The method can reflect the inherent resistance level of the sugarcane identification materials more objectively, and minimize the interaction of environment to ensure more accurate identification result.

Description

Technical field: [0001] The invention belongs to the technical field of identification of plant disease resistance, in particular to a method for identification of sugarcane resistance to smut. Background technique: [0002] Sugarcane smut is one of the major sugarcane diseases in the world. Breeding new varieties resistant to smut is the most economical, environmentally friendly and effective measure to prevent and control the disease. Resistance to smut is an indispensable part of sugarcane variety breeding. The target of smut resistance identification and screening should be carried out in the breeding process. The method of smut resistance identification affects the effectiveness of disease-resistant breeding. So far, domestic sugarcane varieties have been identified for smut resistance by manual inoculation and field planting identification methods. For the disease rate, the fixed 1-9 classification method is used to classify the resistance. After harvesting, the peren...
