Problems solved by technology

[0009] ① Poor repeatability: use Na 2 SO 4 When a solution, acetonitrile, formic acid, etc. are used as denaturants, excessive protein denaturation often occurs, the serum becomes viscous, and the extraction column is easily blocked, thereby affecting the subsequent process; when the C18 extraction column is combined with concentrated sulfuric acid-silica gel column, some organochlorine pesticides It will react with concentrated sulfuric acid and affect the recovery rate; in addition, the concentrated sulfuric acid-silica gel column needs to be prepared immediately, and the concentrated sulfuric acid and silica gel must be fully mixed during the configuration process. The ratio and mixing degree of sulfuric acid and silica gel are consistent
These factors can lead to poor reproducibility
[0010] ②The operation process is cumbersome: in the pretreatment process, the silica gel should be dried overnight at 300°C; the glass container equipped with a mixture of concentrated sulfuric acid and silica gel should be washed with acetone and then dried at 300°C overnight; to ensure that the concentrated sulfuric acid and silica gel are fully mixed requires constant Stirring and mixing; these processes increase the workload of the pretreatment process and consume a lot of time

Abstract

The invention discloses a pretreatment method for assessing persistent organic pollutant level of serum. The pretreatment method comprises four steps, namely (1) protein denaturation, (2) extraction, (3) elution and (4) volume fixation, wherein in the protein denaturation, the protein denaturant is mixed solution of 1-propanol and ultrapure water, and the volume ratio of the 1-propanol to the ultrapure water is 15:85; in the extraction, a C18 extraction column and an alkali aluminum column are used together; in the elution, the eluent is mixed solution of n-hexane and dichloromethane, and thevolume ratio of the n-hexane to the dichloromethane is 80:20; and in the volume fixation, the eluent containing the detected substances is blown with nitrogen, and then the volume is fixed by using isooctane. The method is simple and convenient, has low sample demand, meets the requirement of large-scale epidemiological investigation of crowds, and has good application value.

Description

【Technical field】 [0001] The invention relates to a method for determining the content of a certain substance in a sample, in particular to a pretreatment method for determining the level of persistent organic pollutants in umbilical cord serum by using a gas chromatography electron capture detector. 【Background technique】 [0002] Determining the content of a certain substance in biological samples is the most objective and direct way to assess the exposure of the population, but in the actual operation of epidemiological investigations (especially children) the number of biological samples is not large. Therefore, in epidemiological investigations, it is necessary to establish a simple, convenient, and small-sample evaluation and measurement method. [0003] PCBs and organochlorine pesticides are persistent organic pollutants. They are persistent and fat-soluble. They mainly enter the human body through the food chain, and can also enter the fetus through the placenta. An...
