Problems solved by technology

However, herbicide-resistant rapeseed has not yet been approved for commercial planting in my country. There are two main reasons: (1) Herbicide-resistant rapeseed is generally a genetically modified crop, and commercial production must undergo strict safety evaluation
(2) So far, domestic rapeseed herbicide resistance genes mainly come from abroad, and resistance genes are protected by international intellectual property rights. Once they are commercially planted, they need to pay expensive patent fees, which will greatly increase the production cost of rapeseed, which violates the principle of planting resistance The original intention of herbicide crops
Therefore, the rape-resistant varieties bred or transformed with imidazolinone herbicide-resistant rapeseed new germplasm M9 cannot be popularized and applied in the main production areas of winter rapeseed in my country, which greatly limits the application value of rapeseed imidazolinone-resistant traits

Embodiment

[0025] A: EMS concentration gradient test: In the autumn of 2007, the male parent N131 of the new double-low hybrid rapeseed variety Ningza 15 was selected as the material (Pu Huiming et al., Ningza 15, a new oil-vegetable dual-purpose variety of Brassica napus (B. napus) Breeding, Jiangsu Agricultural Science Journal, 2010, 26(6): 1432~1434), prepared 5 concentrations of 0.2%, 0.4%, 0.6%, 0.8% and 1.0% with pH 7.0, 0.1M phosphate buffer EMS solution, with water as a control, EMS concentration gradient test. Soak the test seeds in clean water for 12 hours, absorb the water on the surface of the seeds with absorbent paper, and perform EMS seed soaking treatment. After 8 hours of treatment, the seeds are rinsed with running water for 4 hours. The treated seeds are neatly arranged in a petri dish covered with filter paper, 100 grains per dish , repeated 3 times, placed in an artificial climate incubator, germinated under 25 degrees of light, and counted the germination rate after...

Abstract

The invention provides a herbicide-resistant rape directive breeding method based on acetolactate synthase (ALS) target esterase, which belongs to a plant trait breeding method. The method comprises the following steps of selecting materials needed by production of rape or breeding of the rape, processing seeds with ethylmethane sulfonate (EMS), mutagenizing the seeds to be isolated and propagated to a M2 generation, and directionally screening herbicide-resistant mutant characters in a large group under the selection pressure of the herbicide adopting the ALS as the target esterase. In order to increase the probability for acquiring the mutant characters, the directive screening can be continuously conducted in multiple years, or conducted in multiple places in the same year or conducted in multiple places in multiple years until the needed mutant material is screened out.

Description

technical field [0001] The invention relates to a herbicide-resistant rapeseed breeding method, in particular to the selection of rapeseed materials, a seed EMS treatment method and the directional screening of herbicide selective pressure resistance traits, belonging to a plant trait breeding method. Background technique [0002] Weeds in farmland seriously affect the yield and quality of rapeseed, and have become one of the major biological disasters in rapeseed production. The promotion and application of herbicide-resistant rapeseed varieties and the use of chemical herbicides for weed control are not only fast, effective, but also economical and practical. Compared with traditional rapeseed, herbicide-resistant rapeseed has simple and efficient farming operations. Growers only need to spray herbicides once or twice at the seedling stage to effectively kill field weeds. Compared with manual weeding, it can save labor , reduce production costs and increase economic return...
