Problems solved by technology

At present, the traditional method of sowing seedlings is mainly used in production to cultivate afforestation seedlings. Due to the diversity of genetic materials, the growth phenotypes of seedlings are different after afforestation, and it is difficult to achieve the goal of high-yield and high-quality groups.

Example Embodiment

[0011] Example 1:

[0012] (1) Selection of good mother trees. In 2005, 10 excellent trees were selected from the 15-year-old Southwestern birch plantation.

[0013] (2) The method of promoting buds at the base of the trunk. In August 2005, weeds and shrubs within a diameter of 5m around the selected mother tree were removed to make the trunk fully exposed to sunlight, and a 6-benzyl adenine (6-BA) solution with a concentration of 5 mg / L was prepared. Use the medical infusion bottle to hang on the trunk at a height of 2m and use the medical infusion tube to maintain the infusion on the cambium at the base of the trunk for 35 days to promote its germination;

[0014] (3) Treatment of explant materials. In October 2005, use scissors to cut the sturdy young leaf buds of 2 to 3 cm in length from the base of the trunk 2m below, and cut off the leaves other than the apical buds as tissue culture explants. A total of 55 leaf buds were collected. The leaf bud explants are first soaked in...

[0017] Example 2:

[0018] (1) Selection of good mother trees. In 2008, 10 excellent trees were selected from the 5-year-old Southwestern birch plantation.

[0019] (2) The method of promoting buds at the base of the trunk. In October 2008, weeds and shrubs within a diameter of 5m around the selected mother tree were removed to make the trunk fully exposed to sunlight, and a 6-benzyl adenine (6-BA) solution with a concentration of 5 mg / L was prepared. , Use a medical infusion bottle to hang on the trunk at a height of 2m and use a medical infusion tube to maintain the infusion on the cambium at the base of the trunk for 32 days to promote its germination;

[0020] (3) Treatment of explant materials. In December 2008, scissors were used to cut 2 to 3 cm long sturdy young leaf buds that sprouted below 2 m from the base of the trunk, and cut off the leaves other than the terminal buds as tissue culture explants. A total of 40 leaf buds were collected. The leaf bud explants are first...

Abstract

The invention discloses an explant collecting and inducing method of leaf buds of betula alnoides, belonging to the technical field of vegetative propagation of plants. The method comprises the following steps of: (1) selecting a betula alnoide superior tree of above five years old as a seed tree; (2) in August-October, clearing weeds and bushes around the seed tree in the diameter range of 5m to ensure that the tree trunk is fully illuminated by sunlight, preparing a 6-benzyladenine solution with the concentration of 5mg/L simultaneously and transfusing the solution into the cambium of the tree trunk base for more than 30 days; (3) in October-December, scissoring and taking 2-3cm long healthy and strong leaf bud produced at 2m below the tree trunk base as the explant, soaking for 20min by using 3% of washing powder, disinfecting the explant for 10-15s by using 75% of alcohol on a super-clean worktable, disinfecting the explant for 7-10min by using 10% of H2O2 and finally washing 5-7 times by using sterile water; and (4) transferring the leaf bud to an induction medium of the explant and culturing for 40-60 days, wherein the induction success rate is more than 50%.

Description

technical field [0001] The invention relates to a method for collecting and inducing leaf bud explants in the tissue culture propagation technology of betula southwest, belonging to the technical field of plant asexual propagation. Background technique [0002] Southwest Betula is an excellent native fast-growing timber tree species of Betulaceae in southern my country. It has strong adaptability and wide uses. It is suitable for cultivating large and medium-diameter timber, but the resources of fine species are very scarce. At present, the traditional method of sowing and raising seedlings is mainly used in production to cultivate afforestation seedlings. Due to the diversity of genetic materials, the seedlings have different growth phenotypes after afforestation, and it is difficult to achieve the goal of high-yield and high-quality groups. The use of plant tissue culture propagation technology can quickly propagate clone seedlings with excellent genes, so as to achieve th...
