Abstract

The invention discloses an explant collecting and inducing method of leaf buds of betula alnoides, belonging to the technical field of vegetative propagation of plants. The method comprises the following steps of: (1) selecting a betula alnoide superior tree of above five years old as a seed tree; (2) in August-October, clearing weeds and bushes around the seed tree in the diameter range of 5m to ensure that the tree trunk is fully illuminated by sunlight, preparing a 6-benzyladenine solution with the concentration of 5mg / L simultaneously and transfusing the solution into the cambium of the tree trunk base for more than 30 days; (3) in October-December, scissoring and taking 2-3cm long healthy and strong leaf bud produced at 2m below the tree trunk base as the explant, soaking for 20min by using 3% of washing powder, disinfecting the explant for 10-15s by using 75% of alcohol on a super-clean worktable, disinfecting the explant for 7-10min by using 10% of H2O2 and finally washing 5-7 times by using sterile water; and (4) transferring the leaf bud to an induction medium of the explant and culturing for 40-60 days, wherein the induction success rate is more than 50%.
