The method comprises the following steps: respectively preparing a cordycep fermenting bacterial liquid and a rainbow conk fermenting bacterial liquid; preparing an edible mushroom enzyme fermenting liquid of fruits and vegetables; sequentially and continuously fermenting the cordycep fermenting bacterial liquid and the rainbow conk fermenting bacterial liquid in the edible mushroom enzyme; mechanically breaking and extruding, and collecting the fermenting liquid. According to the preparation method provided by the invention, by combining actual production of the enzyme, two steps of fermentation by the cordyceps fermenting bacterial liquid and the rainbow conk fermenting bacterial liquid are added in the fermenting process, and cells are mechanically and forcibly crushed to facilitate overflow of the SOD from cells from inside to outside.
