Problems solved by technology

Although the comparison file can construct inbred lines, in most cases, the proportion of hermaphrodites in dioecious shellfish is relatively low, and some special species are even difficult to occur, so the construction of inbred lines still faces many diffic

Example Embodiment

[0017] Example 1

[0018] a. Obtaining sperm: In July 2012, the parent shells of the Philippine clams became sexually mature. In the Dalian Zhuanghe Marine Shellfish Nursery, a method of dry in the shade for 3 hours and water stimulation for 0.5 hours was used to stimulate the parents to discharge gametes. When it starts to discharge gametes, grab single male individuals, rinse them with fresh water, and place them in a beaker filled with 100 mL of fresh seawater to continue to discharge sperm to obtain single individual sperm.

[0019] b. Refrigerated sperm: the individual sperm obtained are precipitated and concentrated by a centrifuge, according to Li Chun (Li Chun. Research on ultra-low temperature preservation of marine economic animal sperm [D]. Qingdao: Master's thesis of the Institute of Oceanology, Chinese Academy of Sciences, 2000) The ultra-low temperature cryopreservation technology refrigerates the sperm of Philippine clam, a total of 90 individual individual sperm. [0022] Example 2

[0023] a. Obtaining sperm: In May 2012, the parent shell of the purple mussel was sexually mature. In the key experiment of the Ministry of Agriculture of Dalian Ocean University, the method of drying in the shade for 2 hours and stimulating by running water for 0.4 hours was used to stimulate the parent to discharge gametes. When it starts to discharge gametes, grab single male individuals, rinse them with fresh water, and place them in a beaker filled with 100 mL of fresh seawater to continue to discharge sperm to obtain single individual sperm.

[0027] Example 3

[0028] a. Obtaining sperm: In June 2012, Hong Kong oyster broodstock was sexually mature. At the Zhanjiang Special Animal Experimental Station of the South China Sea Institute of the Chinese Academy of Sciences, the method of cooling to 4℃ for 12 hours, drying in the shade for 3 hours and running water for 0.5 hours was used to stimulate the parents to discharge gametes. When it starts to discharge gametes, it grabs single male individuals, rinses them with fresh water, and places them in a single beaker filled with 500 mL of fresh sea water to continue to discharge sperm to obtain single individual sperm.

[0029] b. Refrigerated sperm: The individual sperm obtained is precipitated and concentrated by a centrifuge. According to the ultra-low temperature cryopreservation technology of Li Yongmei (Patent Publication Number: CN103141472A), Hong Kong oyster sperm is frozen, and a total of 80 individual sperm are frozen. At the same time, each male individual was num...

Abstract

The invention discloses a new method for building inbred lines of dioecious bivalve molluscs on a large scale. The new method comprises the following steps of acquiring sperms of a monosome; freezing the sperms at ultralow temperature; collecting eggs of a female monosome converted from a male monosome by utilizing a sex reversal characteristic of molluscs on time; activating the frozen sperms, fertilizing with the eggs of the same monosome and incubating to obtain offspring of the same monosome, namely successfully building an inbred line. Different from the traditional habit of building an inbred line by screening spatial hermaphroditic monosomes from dioecious molluscs, the new method has the advantages that the inbred lines of various dioecious bivalve molluscs are built on a large scale, the gene purification speed of the dioecious molluscs is improved, a variety breeding cycle is shortened, and a solid foundation is laid for building pure lines (new varieties); no heterologous gene is imported, and the new method has the advantages of simple operation, high practicability, convenience for promotion and the like.

Description

Technical field: [0001] The invention belongs to the technical field of shellfish seed breeding in marine agriculture, and specifically relates to a new method for large-scale construction of dioecious bivalve inbred lines. Background technique: [0002] Most bivalve molluscs are dioecious, such as clams, cockles, clams, mussels, oysters, etc.; a few species are hermaphrodite, such as bay scallops. Under normal circumstances, an individual of the hermaphrodite shellfish will only produce a single sex. After the gonads mature, an individual can only produce one kind of gametes, namely eggs or sperm; each individual of the hermaphrodite shellfish will produce two at the same time during the development of the gonads. Types of gametes, ie eggs and sperm. [0003] Shellfish often undergo sexual reversal. The so-called sexual reversal refers to the fact that there are both male and female reproductive organs in the shellfish, but they generally exhibit one kind, and sometimes, the othe...
