Embodiment 1

[0018] like figure 1 As shown, one end of the PVC backing 1 is pasted with a sample pad 2 and a nitrocellulose membrane 3 in turn, and the other end is stuck with a water-absorbing pad 4, and the nitrocellulose membrane 3 is coated with ampicillin-BSA as a detection line 5, and goat anti-mouse IgG served as control line 6. The matching microwell plate is coated with anti-β-lactam antibiotic cluster-specific antibody-colloidal gold marker and mouse negative serum-colloidal gold marker.

[0019] Prepare as follows:

[0020] (1) Preparation of ampicillin-BSA conjugate: Dissolve ampicillin and EDC in MES (0.01mol / L, pH 5) at a molar ratio of 1:3, stir and activate at room temperature for 1 hour, and then add the activated substance dropwise to dissolve In BSA-containing HEPES (0.01mol / L, pH 7), the molar ratio of BSA and ampicillin was 1:50, reacted for 4 hours, and dialyzed with normal saline at 4°C for 3 days to form ampicillin-BSA conjugates;

Abstract

The invention discloses a receptor-based immunochromatography test strip for detecting Beta-lactam antibiotics and a preparation method of the test strip, which belong to the technical field of immunological detection. The test strip comprises a PVC (polyvinyl chloride) back lining, wherein the front end of the PVC back lining is provided with a sample pad which is connected with the front end of a cellulose nitrate membrane; the rear end of the cellulose nitrate membrane is connected with a water absorption pad; a combined microporous board is enveloped with a mixture of an anti-Beta-lactam antibiotic receptor-colloidal gold marker and a mouse negative serum-colloidal gold marker; the cellulose nitrate membrane is sequentially enveloped with an ampicillin-BSA (Bovine Serum Albumin)) detection line and a goat anti mouse IgG (Intravenous Gamma Globulin) control line. By virtue of the immunochromatography suitable for on-site detection, the multi-residue detection on at least fifteen Beta-lactam antibiotics can be achieved; the detection is fast and only needs 5-10 minutes; the test strip is convenient to carry, suitable for on-site detection, and simple and convenient to operate without needing professionals.

Description

technical field [0001] The invention relates to a receptor-based immunochromatographic test strip for detecting β-lactam antibiotics and a preparation method thereof, belonging to the technical field of immunological detection. Background technique [0002] Because there are many kinds of veterinary drugs, there are also many kinds of β-lactam antibiotics, and the single residue detection is far from being able to satisfy the good detection of drug residues caused by the abuse of veterinary drugs. Using antigen-antibody reaction can realize the detection of a small part of β-lactam antibiotics, but it is still difficult to meet the needs of multi-residue detection. Existing methods that can achieve multi-residue detection are mainly instrumental analysis, including high performance liquid chromatography, liquid chromatography-mass spectrometry (HPLC-MS), gas chromatography-mass spectrometry (GC-MS), and TTC methods. However, these detection methods require expensive instrum...
