Problems solved by technology

However, artificial synthetic materials have biocompatibility problems to varying degrees, which will cause aseptic inflammation caused by physical stimulation and sequelae caused by chronic rejection.
[0007] The source of autologous cartilage is limited, and it is easy to cause defects in the donor site; allogeneic cartilage is widely used, but it may cause immune rejection or cross-infection; periosteum transplantation has the defect of unstable effect

Embodiment 1

[0034] Embodiment 1, the preparation of biotype cartilage repair material

[0035] 1. Collect the costal cartilage of adult pigs that have just been slaughtered from slaughterhouses under standardized management, try to avoid contact with pollutants, and freeze them immediately after collection.

[0036] 2. Thaw the costal cartilage obtained in step 1 and wash it thoroughly, cut off the muscle tissue, and remove the periosteum (for the results of HE staining at this time, see figure 1 ).

[0037] 3. Decellularization treatment

[0038] Treat the costal cartilage by immersion in a surfactant solution.

[0039] The purpose of this step is to destroy the cell membrane structure and break up the cells for dissolution.

[0040] Specific steps: use 0.5g / 100ml TritonX-100 aqueous solution to soak for 15 hours at 8±2°C, and then wash with water.

[0041] 4. The first alkali treatment

[0042] Use alkaline solution to soak the product of step 3.

Embodiment 2

[0056] Embodiment 2, the preparation of biotype cartilage repair material

[0057] 1. Collect the articular cartilage of adult pigs that have just been slaughtered from slaughterhouses under standardized management, avoid contact with pollutants as much as possible, and freeze them immediately after collection.

[0058] 2. The articular cartilage obtained in step 1 is thawed and fully cleaned, the muscle tissue is cut off, and the periosteum is removed.

[0059] 3. Decellularization treatment

[0060] Treatment of articular cartilage by soaking in a surfactant solution.

[0061] Specific steps: Soak with 1g / 100ml Tween-80 aqueous solution at 23±2°C for 5 hours, then wash with water.

[0062] 4. The first alkali treatment

[0063] Use alkaline solution to soak the product of step 3.

[0064] Specific steps: use 0.5M KOH aqueous solution, soak at 23±2°C for 30 minutes, and then wash with water.

[0065] 5. Degreasing treatment

[0066] Soak the product of step 4 with a sur...

Embodiment 3

[0074] Embodiment 3, the preparation of biological cartilage repair material

[0075] 1. Collect the costal cartilage of adult cattle that have just been slaughtered from slaughterhouses under standardized management, avoid contact with pollutants as much as possible, and freeze and store immediately after collection.

[0076] 2. Thaw the costal cartilage obtained in step 1 and fully wash it, cut off the muscle tissue, and remove the periosteum.

[0077] 3. Decellularization treatment

[0078] Treat the costal cartilage by immersion in a surfactant solution.

[0079] Specific steps: Soak in 3g / 100ml Tween-40 aqueous solution at 2±2°C for 168 hours, then wash with water.

[0080] 4. The first alkali treatment

[0081] Use alkaline solution to soak the product of step 3.

[0082] Specific steps: use 0.02MCa(OH) 2 Aqueous solution, soak at 2±2°C for 180min, then wash with water.

Abstract

The invention discloses a biological type cartilage repair material and its preparation method. The method for preparing the biological type cartilage repair material comprises the following steps: (1) a surfactant solution is used to immerse and process isolated animal cartilage; (2) aqueous alkali is used to immerse and process a product obtained from the step (1); (3) a surfactant solution is used to immerse and process a product obtained from the step (2); (4) aqueous alkali is used to immerse and process a product obtained from the step (3); (5) an irradiation protective reagent solution with pH of 5.0-9.0 is used to immerse and process a product obtained from the step (4); and (6) a product obtained from the step (5) undergoes freeze drying and irradiation sterilization successively so as to obtain the cartilage repair material. The biological type cartilage repair material provided by the invention has advantages of no immunological reaction and good biocompatibility and can meet mechanical requirements of repaired tissues.

Description

technical field [0001] The invention belongs to the field of medical devices, and in particular relates to a biological cartilage repair material for repairing bone tissue in surgery and a preparation method thereof. Background technique [0002] In modern surgical operations, it is often necessary to repair some defective cartilage tissues, such as bone injury repair, articular surface repair, etc. In order to meet the needs of these repairs, a variety of materials and solutions have been clinically used for the repair of cartilage injuries, but at present, there are varying degrees of application defects. [0003] An ideal scaffold material should have the following characteristics: ① good tissue compatibility, non-toxic, non-teratogenic; ② three-dimensional structure; ③ biodegradable; ④ good cell interface; Mechanical strength. [0004] Restorative materials currently used clinically include the following categories: [0005] Natural biological materials: mainly includ...
