Embodiment 1

[0034] Embodiment 1, the acquisition of transgenic MaOFP1 tomato

[0035] 1. Acquisition of the gene MaOFP1

[0036] MATCHMAKER from Clontech TM GAL4Two-Hybrid System 3 kit, using banana MuMADS1 as bait, constructed a cDNA library of banana fruit 2 days after harvest, and transformed yeast AH109 by co-transformation method. Select the blue colonies, extract the yeast plasmid DNA as a template, use the long-distance PCR primers in the kit (P1: 5′-CTATTCGATGATGAAGATACCCCACCAAACCC-3′ and P2: 5′-GTGAACTTGGCGGGGTTTTTCAGTATCTACGATT-3′ and the Advantage 2PCR Polymerase Mix (Clontech, Cat. No. 639201) amplification, the obtained PCR product is sequenced, and full-length primers are designed after bioinformatics analysis, and the amplified nucleotide sequence is SEQ ID No. 1 in the sequence table, The gene shown by the nucleotide sequence is named MaOFP1, the protein it encodes is MaOFP1, and the amino acid sequence of the protein is SEQ ID No. 2 in the sequence listing.

Embodiment 2

[0053] Example 2, Functional Research of MaOFP1

[0054] 1. Phenotype of MaOFP1-transformed tomato

[0055] Sow T2 generation transformed MaOFP1 tomato (MaOFP1), T2 generation transformed empty vector tomato and untransformed wild-type tomato (control) numbered L3, L5, L6, L7, and sow the tomato seeds of the above strains in the seedling pots, at 25 ℃, light 3000lux, culture under 8h / d, and grow for about 2 months. When the plant grows vigorously and the height of the plant is 15-20cm, transfer the seedlings to the field, and observe the fruit at the red maturity stage. There were 20 strains for each strain, and the experiment was repeated three times, and the results were average ± standard deviation.

[0056] The result is as image 3 As shown, where A is the longitudinal section of red ripe fruit; B is the regulation of MaOFP1 on the weight of red ripe fruit, C and D are the regulation of MaOFP1 on the thickness of red ripe fruit peel and pulp, E is the effect of MaOFP1 o...

Abstract

The invention discloses a novel gene MaOFP1 capable of improving the quality of a plant fruit and relates to a related application of the MaOFP1 gene in improving the quality of the plant fruit. The invention provides a method for cultivating a transgenic plant. In order to inoculate the MaOFP1 gene to a target plant to obtain a transgenic plant having at least one of the following characteristics (1)-(4): (1) the weight of the transgenic plant fruit is greater than that of the target plant fruit; (2) the thickness of the transgenic plant fruit flesh is greater than that of the target plant fruit flesh; (3) the thickness of the transgenic plant fruit peel is greater than that of the target plant fruit peel; and (4) the content of soluble solids in the transgenic plant fruit is greater than that in the target plant fruit, the nucleotide sequence of the MaOFP1 gene is SEQ ID No. 1 in sequence table or nucleotide at the 1st-435th positions from 5' terminal of the SEQ ID No. 1 in sequence table; the amino acid sequence of an encoding product MaOFP1 protein of the MaOFP1 gene is SEQ ID No. 2 in the sequence table. Experiments show that the MaOFP1 gene of a banana fruit is inoculated to a wild-type tomato to obtain the transgenic plant with improved quality and a foundation is laid for plant breeding.

Description

technical field [0001] The present invention relates to the field of biotechnology, in particular to a gene for improving plant fruit quality, its encoded product and application. Background technique [0002] OFP (ovate family protein, OFP) (ovate family protein gene) is a class of genes that exist only in plants. The protein encoded by this class of genes has a nucleoid localization signal and a conserved 70 amino acids of unknown function. Domain of Unknown Function 623 (DUF623) domain (Liu et al., 2002), which was later renamed the OVATE domain (Hackbusch et al., 2005). The OVATE gene was first isolated and identified in tomato as an important regulator of fruit shape, which was caused by the early appearance of the terminator of the OVATE gene during translation, which led to the transformation of tomato fruit from round to pear shape (Liu et al., 2002). Later, the AtOFPs gene family with a conserved OVATE domain was also discovered in Arabidopsis and shown to regulate...
